目的:探讨龙柴方提取液及其含药血清对CCL4诱导肝细胞损伤保护作用。方法:培养L-02型肝细胞,12h后用四氯化碳(CCL4)体外诱导肝细胞损伤,分为正常模型对照组、7组不同浓度龙柴方药液组和正常对照组,继续培养12h,MTT检测各组细胞存活力;同时,诱导肝细胞损伤模型制成后,分为CCL4模型组、正常血清对照组、龙柴方血清组、益肝灵血清组,继续培养12h,MTT检测细胞各组存活力。结果:CCL4模型组及各药液组细胞OD值均低于正常对照组(P<0.01),除龙柴方最高浓度组低于模型组外(P>0.05),其余6药液组细胞OD值均高于CCL4模型组;龙柴方血清组及益肝灵血清组细胞OD值明显高于CCL4模型组和正常血清组(P<0.05),以龙柴方血清组效果为优。结论:龙柴方提取液具有抗肝细胞损伤和促进肝细胞生长的药理作用。
Objective: To investigate the protective effect of Longchaifang extract and its drug-containing serum on CCL4-induced hepatocyte injury. METHODS: L-02 hepatocytes were cultured. After 12 hours, hepatocyte injury was induced with carbon tetrachloride (CCL4) in vitro. The hepatocytes were divided into normal model control group, 7 different concentrations of Longchaifang liquid group and normal control group, and culture was continued. At 12h, MTT assayed the viability of cells in each group; at the same time, after induction hepatocyte injury model was made, they were divided into CCL4 model group, normal serum control group, Longchaifang serum group, Yiganling serum group, and cultured for 12 hours, MTT assay. Results: The OD values in the CCL4 model group and each drug solution group were lower than those in the normal control group (P<0.01), except for the Longchaifang highest concentration group was lower than the model group (P>0.05), and the remaining 6 drug solution group cells OD The values of OD values in Longchaifang serogroup and Yiganling serogroup were significantly higher than those in CCL4 model group and normal sera group (P<0.05). The effect of Longchaifang serogroup was superior. Conclusion: Longchaifang extract has the pharmacological effects of hepatocyte injury and promotion of hepatocyte growth.
