目的探讨脑源性神经营养因子(BDNF)和神经营养因子3(NT-3)在扬子鳄大脑胚后发育中的时序表达。方法用免疫组织化学染色和Western blotting法观察BDNF和NT-3在0岁、1岁和2岁的扬子鳄大脑皮质中的定位和含量变化。结果免疫组织化学显示,0岁扬子鳄大脑皮质区内4层均可见BDNF阳性细胞,随着年龄增加,BDNF阳性细胞数量逐渐增多,到2岁时BDNF阳性细胞胞体增大,突起增多,主要集中在细胞层和外网状层移行区。这与Western blotting检测结果一致。0岁扬子鳄大脑皮质区内可见NT-3阳性细胞,主要集中在细胞层和外网状层。到1岁时,NT-3阳性细胞逐渐增多,在外网状层可见大量密集的阳性细胞。2岁时,外侧的分子层中NT-3阳性细胞增多,且细胞突起随着年龄增加而出现增多。这一结果与Western blotting检测一致。结论 BDNF和NT-3参与了扬子鳄胚后2年内大脑皮质区神经细胞的生长发育。
Objective To investigate the temporal expression of brain-derived neurotrophic factor (BDNF) and neurotrophic factor 3 (NT-3) in the embryonic brain of Alligator sinensis. Methods The localization and content of BDNF and NT-3 in cerebral cortex of Chinese alligator (crocodile) at 0, 1 and 2 years old were observed by immunohistochemistry and Western blotting. Results Immunohistochemistry showed that BDNF positive cells were found in all four layers of cortex of 0-year-old Chinese crocodile. BDNF positive cells gradually increased with age, and the number of BDNF positive cells increased and the number of protrusions increased at the age of 2, Cell layer and outer reticular layer transition zone. This is consistent with Western blotting results. NT-3 positive cells were found in the cortex of 0-year-old Chinese crocodile, which were mainly concentrated in the cytoplasm and outer reticular layer. By the age of 1, NT-3 positive cells gradually increased and a large number of dense positive cells were visible on the outer reticular layer. At age 2, NT-3 positive cells increased in the outer molecular layers, and the number of cell protrusions increased with age. This result is consistent with Western blotting. Conclusion BDNF and NT-3 are involved in the growth and development of nerve cells in cerebral cortex within 2 years after allogeneic embryogenesis.
