目的:探讨ABCG2、CD133、CK20、CyclinB1和DNA-PKcs在SGC-7901和MGC-803细胞SP(side population)亚群中的表达,探讨其为胃癌干细胞(cancer stemcell)标志的可行性。方法:用Hoe-chest33342染色检测胃癌细胞SGC-7901和MGC-803中SP细胞的比例;通过荧光免疫细胞化学染色比较ABCG2、CD133、CK20、Cyclin B1和DNA-PKcs在SP细胞与非SP细胞中表达的差异。结果:经Hoechst33342和荧光免疫细胞化学染色显示,SGC-7901细胞中SP细胞占3.8%;SP细胞中ABCG2和CK20表达阳性的细胞比率显著低于非SP细胞,P<0.05;CD133、Cyclin B1和DNA-PKcs在SP细胞和非SP细胞中的表达差异无统计学意义,P>0.05。经Hoechst33342和荧光免疫细胞化学染色显示,MGC-803细胞中SP细胞占2.9%;SP细胞中Cyclin B1的阳性表达率显著高于非SP细胞中的(P<0.05),但ABCG2、CD133、CK20及DNA-PKcs的阳性表达率显著低于非SP细胞中的,P<0.05。结论:CK20-/Cyclin B1+或CD133+可能是胃癌干细胞的特异性分子标志,而单项ABCG2、CD133、Cyclin B1及DNA-PKcs在鉴定胃癌干细胞时无特异性。
OBJECTIVE: To investigate the expression of ABCG2, CD133, CK20, CyclinB1 and DNA-PKcs in the side population of SGC-7901 and MGC-803 cells and to explore their feasibility as markers of cancer stemcell. Methods: The proportion of SP cells in gastric cancer cells SGC-7901 and MGC-803 was detected by Hoe-chest33342 staining. ABCG2, CD133, CK20, Cyclin B1 and DNA-PKcs were compared in SP cells and non-SP cells by fluorescence immunocytochemistry Differences in expression. Results: Hoechst33342 and fluorescent immunocytochemical staining showed that the percentage of SP cells in SGC-7901 cells was 3.8%. The percentage of cells with positive expression of ABCG2 and CK20 in SP cells was significantly lower than that in non-SP cells (P <0.05). CD133, Cyclin B1 The expression of DNA-PKcs in SP cells and non-SP cells showed no significant difference (P> 0.05). The number of SP cells in MGC-803 cells was 2.9% by Hoechst33342 staining and fluorescent immunocytochemical staining. The positive rate of Cyclin B1 in SP cells was significantly higher than that in non-SP cells (P <0.05), but ABCG2, CD133, CK20 And DNA-PKcs were significantly lower than those in non-SP cells (P <0.05). CONCLUSIONS: CK20- / Cyclin B1 + or CD133 + may be specific molecular markers of gastric cancer stem cells. Individual ABCG2, CD133, Cyclin B1 and DNA-PKcs are not specific in the identification of gastric cancer stem cells.
