目的研究生长相关蛋白-43(growth associated protein-43,GAP-43)在新生鼠缺氧缺血损伤后心肌细胞中的差异表达及其临床意义。方法取42只7日龄Wistar大鼠随机分为正常对照组和缺氧缺血组。大鼠分别于缺氧缺血后2d,3d,7d断头处死取心肌,应用免疫组化法检测心肌细胞上GAP-43的表达。结果缺氧缺血组GAP-43的表达较对照组明显增加,在缺氧缺血后2d至7d差异有显著意义(P<0.05)。结论缺氧缺血大鼠心肌GAP-43表达呈时间依赖性变化,与心肌细胞损害密切相关。
Objective To investigate the differential expression of growth associated protein-43 (GAP-43) in neonatal rat cardiomyocytes after hypoxic-ischemic insult and its clinical significance. Methods Forty-two 7-day-old Wistar rats were randomly divided into normal control group and hypoxic-ischemic group. Rats were sacrificed on the 2nd, 3rd, and 7th day after hypoxia-ischemia, respectively. The expression of GAP-43 in cardiomyocytes was detected by immunohistochemistry. Results The expression of GAP-43 in hypoxic-ischemic group was significantly higher than that in control group, and there was significant difference between 2 and 7 days after hypoxia-ischemia (P <0.05). Conclusion The expression of GAP-43 in hypoxic-ischemic myocardium changes in a time-dependent manner, which is closely related to the damage of myocardial cells.
