目的:观察酪氨酸磷酸酶(protein tyrosine phosphotases,PTP)抑制剂过钒酸钠(sodium Pervanadate,Per)对去除EPO状态下BET-2细胞存活、增殖、线粒体膜电位改变与细胞凋亡的影响。方法:应用MTT法、流式细胞仪等方法观察Per对EPO依赖的小鼠源性髓系白血病细胞株BET-2细胞在无EPO状态下细胞存活、增殖、细胞线粒体膜电位改变、细胞凋亡的影响。结果:BET-2细胞对EPO有良好的依赖性,在无EPO状态下BET-2细胞增殖停滞、细胞线粒体膜电位下降,并出现细胞凋亡;适当浓度Per(1~31.25μmol/L)能促进无EPO状态下BET-2细胞存活、增殖,维持细胞线粒体膜电位,减少细胞凋亡。结论:通过Per等调控PTP活性可能促进EPO依赖的BET-2细胞在EPO缺乏状态下的存活、增殖。
OBJECTIVE: To observe the effect of sodium pervanadate (Per), a tyrosine phosphotase (PTP) inhibitor, on the survival, proliferation, mitochondrial membrane potential and cell apoptosis of BET-2 cells in vitro . Methods: MTT and flow cytometry were used to observe the effects of Per on cell survival, proliferation, mitochondrial membrane potential and cell apoptosis in EPO-dependent mouse MLF cell line BET-2 without EPO Impact. RESULTS: The BET-2 cells had a good dependence on EPO. The proliferation of BET-2 cells was arrested in the absence of EPO, and the mitochondrial membrane potential of cells was decreased and apoptosis was observed. Per (1-31.25 μmol / L) Promote the survival and proliferation of BET-2 cells without EPO, maintain mitochondrial membrane potential and reduce apoptosis. Conclusion: The regulation of PTP activity by Per et al. May promote the survival and proliferation of EPO-dependent BET-2 cells in the absence of EPO.
