用亲和层析法从3~5月龄引产胎儿肌肉组织中提纯AFP。用纯化AFP免疫小鼠,取其脾脏按匀浆透析法制备AFP抗原特异转移因子(SP-TF)。密度离心法分离鼠脾淋巴细胞,体外用~2H-亮氨酸参入法,从翻译水平观察SP-TF对AFP抑制鼠脾淋巴细胞生长的拮抗效应。结果表明,所提纯AFP有非常显著抑制鼠脾淋巴细胞蛋白质合成作用(P<0.001)。SP-TF和AFP抗体能非常显著拮抗AFP抑制鼠脾淋巴细胞~2H-亮氨酸参入效应(P均<0.001),两者对AFP的拮抗作用无显著差异(P>0.05),而正常小鼠脾制备的转移因子(N-TF)无此拮抗效应。SP-TF对AFP的拮抗效应并非是SP-TF本身对淋巴细胞作用的结果。
Purification of AFP from 3 to 5-month-old fetal muscle tissue by affinity chromatography. Mice were immunized with purified AFP and their spleens were prepared by homogenization dialysis to prepare AFP antigen-specific transfer factor (SP-TF). The splenic lymphocytes were isolated by density centrifugation, and the effect of SP-TF on the inhibition of the growth of murine splenic lymphocytes by AFP was observed from the translational level using ~ 2H-leucine incorporation method. The results showed that the purified AFP had a significant inhibitory effect on the protein synthesis of rat splenic lymphocytes (P <0.001). SP-TF and AFP antibodies could significantly antagonize the effect of AFP on -2H-leucine incorporation in splenic lymphocytes of mice (all P <0.001), but no significant difference was found between the two groups (P> 0.05) Transfer factor (N-TF) prepared from rat spleens had no such antagonistic effect. The antagonistic effect of SP-TF on AFP is not the result of SP-TF itself acting on lymphocytes.
