目的:研究白花蛇舌草总黄酮(FOD)对人体分化胃腺癌BGC-823细胞增殖、凋亡的影响及其可能的作用机制。方法:MTT法检测FOD对BGC-823细胞增殖的影响;流式细胞术测定细胞周期与凋亡的变化;Western-blot法评价埃茲蛋白(Ezrin)、细胞间黏附分子1(ICAM-1)的表达情况。结果:FOD分别作用BGC-823细胞24h、48h后半数抑制浓度(IC50)依次为12.58、8.43μg/mL;阻滞细胞于G0/G1期(P<0.05)并诱导细胞凋亡(P<0.05);活化的Ezrin表达降低、ICAM-1表达升高。结论:FOD呈时间-剂量依赖性抑制BGC-823细胞增殖,阻滞细胞于G0/G1期,并诱导其凋亡,其诱导凋亡的机制可能与下调Ezrin、上调ICAM-1的表达有关。
Objective: To study the effect of total flavonoids of Hedyotic diffusa (FOD) on the proliferation and apoptosis of human gastric cancer cell line BGC-823 and its possible mechanism. Methods: The effect of FOD on the proliferation of BGC-823 cells was detected by MTT assay. The cell cycle and apoptosis were detected by flow cytometry. The expressions of Ezrin, ICAM-1, The expression of the situation. Results: FOD treated BGC-823 cells at 24h and 48h respectively, the IC50 values were 12.58 and 8.43μg / mL, respectively. The cells were arrested in G0 / G1 phase (P <0.05) ); Activated Ezrin expression decreased, ICAM-1 expression increased. CONCLUSION: FOD inhibits the proliferation of BGC-823 cells in a time-and dose-dependent manner, arrests cells in G0 / G1 phase and induces apoptosis. The mechanism of FOD induction may be related to down-regulation of Ezrin and up-regulation of ICAM-1 expression.
