目的建立同时测定槲寄生药材中高圣草素、槲皮素-3,3′-二甲醚、商陆素和藿香黄酮醇量的反相高效液相色谱法。方法固定相为Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸溶液(40∶60)(A)和乙腈(B)梯度洗脱,体积流量为1.0 mL/min,柱温为30℃,检测波长为280 nm。结果在上述条件下测得高圣草素、槲皮素-3,3′-二甲醚、商陆素和藿香黄酮醇的质量浓度分别在0.041 2~0.824、0.007 6~0.152、0.036 0~0.720、0.003 20~0.064 0 mg/mL时与色谱峰面积之间线性关系良好;平均回收率分别为97.63%(RSD为1.6%),96.4%(RSD为1.9%),97.5%(RSD为1.7%),98.1%(RSD为2.5%)。结论该方法准确、灵敏,可作为槲寄生药材质量控制的一个有效方法。
OBJECTIVE To establish an RP-HPLC method for the simultaneous determination of the contents of homostilbene, quercetin-3,3’-dimethyl ether, mespan and genistein in mistletoe medicinal materials. Methods The mobile phase consisted of a Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) with a gradient of methanol-0.1% phosphoric acid (40:60) (A) and acetonitrile (B) with a volume flow of 1.0 mL / min The column temperature was 30 ℃ and the detection wavelength was 280 nm. Results Under the above conditions, the mass concentrations of emodin, quercetin-3,3’-dimethyl ether, melatonin and genistein were 0.041 2 ~ 0.824, 0.007 6 ~ 0.152, 0.036 0 ~ 0.720,0.003 20 ~ 0.064 0 mg / mL, the average recovery was 97.63% (RSD 1.6%), 96.4% (RSD 1.9%) and 97.5% (RSD 1.7%), 98.1% (RSD 2.5%). Conclusion The method is accurate and sensitive and can be used as an effective method for quality control of mistletoe medicinal materials.
