目的观察胃癌组织中缺氧诱导因子-1α(HIF-1α)、信号传导与转录活化因子3(STAT3)的表达变化及其与患者预后的关系。方法免疫组化染色法检测123例份胃癌根治术切除的胃癌组织和53例份正常胃黏膜组织中的HIF-1α和STAT3。结果胃癌组织中HIF-1α、STAT3的阳性表达率分别为64.2%(79/123)和62.6%(77/123),与正常胃黏膜组织的39.6%(21/53)和43.4%(23/53)相比,P均<0.05。HIF-1α及STAT3的阳性表达与胃癌病理分化程度、肿瘤浸润深度、有无淋巴结转移、TNM分期及有无血管侵袭有关(P均<0.05),HIF-1α、STAT3共同阳性表达率随胃癌TNM分期的升高而升高(r=0.377,P<0.05)。胃癌组织中HIF-1α与STAT3的表达呈正相关(r=0.335,P<0.05)。HIF-1α和STAT3阳性表达者术后5年生存率分别为21.9%和27.4%,阴性表达者分别为51.2%和41.5%,二者相比,P均<0.05。胃癌组织中HIF-1α阳性表达是影响胃癌患者预后的独立危险因素(RR=2.162,P<0.05)。结论胃癌组织中HIF-1α、STAT3的阳性表达率升高,二者的表达呈正相关,HIF-1α、STAT3阳性表达者的预后差,HIF-1α阳性表达和TNM分期对评估胃癌患者的预后有一定价值。
Objective To observe the expression of hypoxia-inducible factor-1α (HIF-1α), signal transducer and activator of transcription 3 (STAT3) in gastric cancer and its relationship with prognosis. Methods Immunohistochemical staining was used to detect HIF-1α and STAT3 in 123 cases of gastric cancer tissues resected by radical gastrectomy and 53 cases of normal gastric mucosa. Results The positive expression rates of HIF-1α and STAT3 in gastric cancer tissues were 64.2% (79/123) and 62.6% (77/123), respectively, and 39.6% (21/53) and 43.4% (23/4%) of normal gastric mucosal tissues. 53) Compared with P<0.05.
