目的通过对中国(未包括香港、澳门特别行政区和台湾地区)脊髓灰质炎(脊灰)实验室网络(Chinese Poliomyelitis Laboratory Network,CPLN)2007年4月~2015年4月连续的细胞敏感性(Cell Sensitivity,CS)监测结果进行分析,评估其在脊灰实验室检测质量控制中的重要作用。方法全国脊灰实验室网络中所有实验室统一使用96孔微量板病毒滴定法在L20B细胞和RD细胞上对三个型别的脊灰病毒的敏感性进行检测。结果从2007~2015年,在CPLN中,有30个省级脊灰实验室已开展CS监测并按时向国家脊灰实验室上报结果,目前西藏由于实验室条件所限尚未开展CS监测。八年间的CS监测结果显示30个省级脊灰实验室的实验室质量控制(LQC)株滴度均在正常范围±0.5 logCCID_(50)/0.1ml波动。结论 2007~2015年CPLN中各个实验室所用的L20B和RD细胞对脊灰病毒敏感性处于正常范围内,细胞系状态稳定,对三个型别的脊灰病毒均敏感。CS监测作为CPLN质量控制的重要指标保证了实验室检测脊灰病毒的敏感性,为从细胞中扩增出低滴度的脊灰病毒提供了保障,对输入脊灰野病毒或疫苗衍生脊灰病毒(VDPV)的早期发现至关重要。
OBJECTIVE: To determine the continuous cell sensitivity of Chinese (excluding Hong Kong, Macau Special Administrative Region and Taiwan) Chinese Poliomyelitis Laboratory Network (CPLN) from April 2007 to April 2015 Sensitivity, CS) monitoring results to assess its important role in the quality control of polio laboratory testing. Methods The sensitivity of three types of poliovirus was detected in L20B cells and RD cells using 96-well microplate titration by all laboratories in the National Polio Laboratory Network. Results From 2007 to 2015, in the CPLN, 30 provincial polio laboratories conducted CS surveillance and reported the results to the National Polio Laboratory on time. Currently, CS monitoring has not been conducted in Tibet due to laboratory conditions. Eight years of CS monitoring results showed that the laboratory quality control (LQC) titers of 30 provincial polio laboratories fluctuated within ± 0.5 log CCID 50 / 0.1 ml in the normal range. CONCLUSIONS: The L20B and RD cells used in laboratories in CPLN from 2007 to 2015 are within the normal range of poliovirus susceptibility. The cell lines are stable and sensitive to all three types of poliovirus. CS monitoring as an important indicator of CPLN quality control to ensure laboratory sensitivity of detection of poliovirus provides a guarantee for the amplification of low-titer poliovirus from cells, and for the input of poliovirus or vaccine-derived poliovirus (VDPV) early detection is crucial.
