目的:探讨细胞凋亡在RSV肺炎发病中的作用以及金欣口服液干预RSV感染的可能机制。方法:体外建立呼吸道合胞病毒A型LONG株攻击体外培养的人胚肺成纤维细胞(HELF)模型,设正常细胞组、病毒组、空白血清组、利巴韦林组和金欣口服液组。采用流式细胞术检测其对感染细胞不同时间点的总凋亡率、早期凋亡率。结果:RSV感染12、24h HELF细胞的总凋亡率、早期凋亡率均低于细胞对照组,且有显著性差异(P<0.05);空白血清组、利巴韦林组、金欣口服液组12、24h的总凋亡率、早期凋亡率均高于病毒对照组,且有显著性差异(P<0.05)。但增加的程度不同,12、24h HELF细胞的总凋亡率和24h早期凋亡率依次排序是空白血清组、利巴韦林组、金欣口服液组;12h早期凋亡率依次排序是利巴韦林组、空白血清组和金欣口服液组。RSV感染48、72、96h HELF细胞的总凋亡率、早期凋亡率均高于细胞对照组,且有显著性差异(P<0.05);空白血清组48、72、96h的总凋亡率均高于病毒对照组(P<0.05)、48h早期凋亡率无变化(P>0.05),72、96h早期凋亡率低于病毒对照组,且有显著性差异(P<0.05);利巴韦林组、金欣口服液组48、72、96h总凋亡率、早期凋亡率均低于病毒对照组,且有显著性差异(P<0.05)。但降低的程度不同,排序依次是金欣口服液组、利巴韦林组、空白血清组(P<0.05)。结论:金欣口服液直接或间接调控细胞凋亡,实现阻止RSV感染早期抑制细胞凋亡、感染后期促进凋亡的作用,从而阻止病毒在细胞内的进一步复制和清除,这可能是金欣口服液抗RSV病毒的作用机制之一。
Objective: To investigate the role of apoptosis in the pathogenesis of RSV pneumonia and the possible mechanism of Jinxin oral intervention in RSV infection. METHODS: Human lung fibroblasts (HELF) models in vitro cultured with a respiratory syncytial virus type A LONG strain were challenged. The normal cell group, virus group, blank serum group, ribavirin group and Jinxin oral solution group were set up. . Flow cytometry was used to detect the total and early apoptosis rates of infected cells at different time points. Results: The total apoptosis rate and early apoptosis rate of HELF cells at 12 and 24 h after infection with RSV were lower than those in the control group (P<0.05); blank serum group, ribavirin group, and Jinxin oral The total apoptotic rate and early apoptotic rate in the fluid group were higher than those in the virus control group at 12, 24 h, and there was a significant difference (P<0.05). However, the degree of increase was different. The total apoptosis rate and 24h early apoptosis rate of HELF cells at 12 and 24 hours were ranked as blank serum group, ribavirin group, and Jinxin oral solution group; Bavelin group, blank serum group and Jinxin oral solution group. The total apoptosis rate and early apoptosis rate of HELF cells infected by RSV were higher than those in the control group at 48, 72, and 96 hours (P<0.05). The total apoptosis rates at 48, 72, and 96 hours in the blank serum group were significant. All of them were higher than the virus control group (P<0.05), and there was no change in the early apoptosis rate at 48 hours (P>0.05). The early apoptosis rate at 72 and 96 hours was lower than that in the virus control group, and there was a significant difference (P<0.05). The total apoptotic rate and early apoptotic rate at 48h, 72h, and 96h in the Bavelin and Jinxin oral solutions groups were lower than those in the virus control group, and there were significant differences (P<0.05). However, the degree of reduction was different, and the order was Jinxin oral solution group, Ribavirin group, and blank serum group (P<0.05). Conclusion: Jinxin Oral Liquid directly or indirectly regulates cell apoptosis, prevents RSV infection from inhibiting apoptosis in the early stage, and promotes apoptosis at the late stage of infection, thereby preventing further replication and clearance of the virus in the cell. This may be due to Jin Xin’s oral administration. Liquid anti-RSV virus one of the mechanisms of action.
