目的 研究汉滩病毒 (HTV)感染对应激基因表达的影响。方法 病毒感染 ,Westernblot,免疫组化 ,免疫荧光双标记 ,激光扫描共聚焦显微镜观察 ,RNA斑点杂交 ,原位杂交。结果 HTV感染细胞中Hsp70高表达 ,病毒感染后Hsp70从细胞浆转位至细胞核 ,病毒感染后不同时间点Hsp70的分布不一 ,间接免疫荧光双标记显示感染细胞的胞浆中NP与Hsp70有共定位。结论 汉滩病毒能直接诱导Hsp70的高表达 ,可能与病毒的装配有关 ,感染细胞中汉滩病毒蛋白与Hsp70共定位 ,并可能形成一种物理复合物
Objective To study the effect of Hantaan virus (HTV) infection on the expression of stress genes. Methods Virus infection, Western blot, immunohistochemistry, immunofluorescence double labeling, laser scanning confocal microscopy, RNA dot blot hybridization, in situ hybridization. Results Hsp70 was highly expressed in HTV infected cells. Hsp70 translocated from the cytoplasm to the nucleus after virus infection. The distribution of Hsp70 was different at different time points after infection with the virus. Indirect immunofluorescence double labeling showed that the cytoplasm of infected cells was co-cultured with Hsp70 Positioning. Conclusion Hantaan virus can directly induce the high expression of Hsp70, which may be related to the assembly of the virus. Hantaan virus proteins co-localize with Hsp70 in infected cells and may form a physical complex
