目的:建立HPLC-PDA法同时测定石榴健胃片中羟基红花黄色素A、鞣花酸、桂皮醛、胡椒碱的含量。方法:采用Phenomenex Gemini C18色谱柱(4.6 mm×250 mm,5μm),以乙腈(A)-0.2%磷酸水溶液(B)为流动相,梯度洗脱,流速为1.0 m L·min~(-1),柱温为30℃,检测波长为403 nm(检测羟基红花黄色素A)和267 nm(检测鞣花酸、桂皮醛、胡椒碱)。结果:羟基红花黄色素A、鞣花酸、桂皮醛、胡椒碱质量浓度分别在1.02~61.37、0.97~57.97、1.18~70.51、1.00~60.29μg·m L~(-1)范围内均呈良好的线性关系,相关系数分别为0.999 7、0.999 9、0.999 9、0.999 9;平均加样回收率(n=6)分别为100.5%、99.7%、100.4%和100.3%,RSD分别为1.2%、1.1%、1.4%和0.98%;供试品溶液在24 h内稳定;含量分别为1.13、0.33、1.08、2.71 mg·g~(-1)。结论:利用HPLC-PDA法建立一种同时测定石榴健胃片中活性成分羟基红花黄色素A、鞣花酸、桂皮醛、胡椒碱含量的方法,该方法简便准确,专属性强,重复性好。经方法学验证,本法可用于石榴健胃片中羟基红花黄色素A、鞣花酸、桂皮醛、胡椒碱的含量测定。
Objective: To establish a HPLC-PDA method for simultaneous determination of hydroxysafflor yellow A, ellagic acid, cinnamaldehyde and piperine in Pomegranate Tablets. METHODS: A gradient elution was carried out on a Phenomenex Gemini C18 column (4.6 mm × 250 mm, 5 μm) using a mobile phase of acetonitrile (A) -0.2% phosphoric acid (B) at a flow rate of 1.0 mL · min -1 ). The column temperature was 30 ℃. The detection wavelength was 403 nm (detection of safflower yellow A) and 267 nm (detection of ellagic acid, cinnamaldehyde and piperine). Results: The concentrations of hydroxysafflor yellow A, ellagic acid, cinnamaldehyde and piperine were all in the range of 1.02 ~ 61.37,0.97 ~ 57.97,1.18 ~ 70.51,1.00 ~ 60.29μg · m L ~ (-1) The correlation coefficients were 0.999 7,0.999 9,0.999 9,0.999 9, respectively. The average recoveries (n = 6) were 100.5%, 99.7%, 100.4% and 100.3%, respectively, with RSDs of 1.2% , 1.1%, 1.4% and 0.98%, respectively. The test solution was stable within 24 h. The contents were 1.13, 0.33, 1.08 and 2.71 mg · g -1, respectively. Conclusion: The HPLC-PDA method was used to establish a method for simultaneous determination of hydroxysafflor yellow A, ellagic acid, cinnamaldehyde and piperine in Pomegranate Stomach Tablet. The method is simple, accurate, specific and reproducible it is good. The methodological validation, the method can be used Pomegranate stomach tablets of hydroxysafflor yellow A, ellagic acid, cinnamic aldehyde, piperine content determination.
