目的:观测 T 淋巴细胞——MT_2细胞对乙型肝炎病毒感染的敏感性,了解病毒感染对淋巴细胞表达 Fas 的影响。方法:体外培养 MT_2细胞,用 HBV DNA 阳性血清直接感染。用酶联免疫法和斑点杂交,PCR 法检测感染后细胞病毒复制与表达状况,用酶联免疫法检测 Fas 抗原表达。结果:MT_2细胞于阳性血清感染后第八天可测到 HBeAg、HBsAg 和HBV DNA,同时出现 Fas 表达信号。结论:MT_2细胞对 HBV 易感,HBV 可上调 MT_2表达 Fas,提示通常在乙型肝炎发病中作为效应细胞的淋巴细胞,亦可能成为凋亡牺牲品。
Objective: To observe the sensitivity of T lymphocyte-MT2 cells to hepatitis B virus infection and to understand the effect of virus infection on the expression of Fas in lymphocytes. Methods: MT 2 cells were cultured in vitro and directly infected with HBV DNA positive serum. Enzyme-linked immunosorbent assay (ELISA) and dot blotting were used to detect the replication and expression of the virus after infection. The Fas antigen expression was detected by enzyme-linked immunosorbent assay. Results: MT2 cells could detect HBeAg, HBsAg and HBV DNA on the eighth day after positive sera infection, and Fas expression signal appeared at the same time. CONCLUSION: MT 2 cells are susceptible to HBV, and HBV can up-regulate the expression of Fas in MT 2, suggesting that lymphocytes, which are usually effector cells in the pathogenesis of hepatitis B, may also be the victims of apoptosis.
