目的 探讨大鼠脑微血管内皮细胞的培养方法。方法 取Wistar大鼠乳鼠脑组织,采用筛网过滤、胶原酶消化、离心等技术获取脑微血管内皮细胞,并进行培养。通过形态学、免疫细胞化学方法进一步鉴定,采用MTT方法测定生长曲线。结果 经形态学、免疫细胞化学方法鉴定所培养的细胞为脑微血管内皮细胞,观察到原代脑微血管内皮细胞有3种表型,细胞呈单层生长,并可传代培养。结论 建立大鼠脑微血管内皮细胞的培养方法可为体外研究脑血管病提供有益帮助。
Objective To investigate the culture method of rat brain microvascular endothelial cells. Methods The brain tissue of Wistar rats was obtained. The brain microvascular endothelial cells were obtained by sieving, collagenase digestion and centrifugation. The morphological and immunocytochemical methods were used to further identify the growth curve by MTT method. Results The morphological and immunocytochemical methods were used to identify the cultured cells as brain microvascular endothelial cells. Three types of primary brain microvascular endothelial cells were observed. The cells were monolayer and subcultured. Conclusion The establishment of rat brain microvascular endothelial cell culture methods for the study of cerebrovascular disease provide useful help.
