目的:考察葛根素对荷人乳腺癌去卵巢裸鼠肿瘤生长、细胞周期及其凋亡的影响。方法:乳腺癌细胞MDA-MB-231接种于去卵巢裸鼠,待肿瘤长至约0.1 cm~3时随机分组,经口服给予葛根素125、25、5 mg·kg~(-1)干预,以肿瘤生长曲线、肿瘤重量、肿瘤细胞流式细胞仪检测等指标考察葛根素对乳腺癌肿瘤细胞的影响。结果:葛根素能明显抑制去卵巢裸鼠乳腺癌的生长,125 mg·kg~(-1)剂量对肿瘤体积和重量的抑制率分别达24.2%和30.3%,并呈一定的量效关系;且能促进乳腺癌细胞的凋亡,抑制细胞周期S期到G_2/M期的转换。结论:葛根素具有抑制荷人乳腺癌去卵巢裸鼠肿瘤生长作用,其机制可能与促进乳腺癌细胞的凋亡、抑制细胞周期S期到G_2/M期的转换有关。
Objective: To investigate the effects of puerarin on tumor growth, cell cycle and apoptosis in nude mice bearing ovariectomized nude mice. Methods: Breast cancer cells MDA-MB-231 were inoculated into ovariectomized nude mice. When the tumors grew to about 0.1 cm ~ 3, they were randomly divided into groups. After oral administration of puerarin 125, 25 and 5 mg · kg -1, The effects of puerarin on breast cancer cells were investigated by tumor growth curve, tumor weight and tumor cell flow cytometry. Results: Puerarin could significantly inhibit the growth of breast cancer in ovariectomized nude mice. The dose of 125 mg · kg -1 could inhibit tumor volume and weight by 24.2% and 30.3%, respectively. And can promote the apoptosis of breast cancer cells, inhibit the cell cycle S phase to G_2 / M phase conversion. CONCLUSION: Puerarin can inhibit tumor growth in nude mice bearing ovariectomized human breast cancer. The mechanism may be related to promoting the apoptosis of breast cancer cells and inhibiting the transition from S phase to G 2 / M phase in breast cancer cells.
