本研究通过对木薯根不同发育时期的酸性转化酶和蔗糖合酶进行m RNA表达量检测,旨在为木薯块根膨大过程中韧皮部卸载路径的阐明提供一定的证据。采用了实时荧光定量PCR技术、△△Ct法对蔗糖代谢酶m RNA相对表达量进行测定。质外体途径的调控酶细胞壁和可溶性酸性转化酶都在须根里表现出了较高的表达量,尤其是细胞壁性酸性转化酶在两种须根里的表达量都显著高于三个发育时期的块根,与之相反,共质体途径的调控酶蔗糖合酶在块根中的表达量则显著高于须根,特别是膨大期的表达量在这些组织中表现出最高水平。因此,细胞壁酸性转化酶和蔗糖合酶分别在须根和块根中的相对高表达,初步证实了须根的质外体和块根的共质体卸载途径的主体地位。
In this study, m RNA expression levels of acid invertase and sucrose synthase in different developmental stages of cassava roots were detected in order to provide some evidence for elucidation of phloem unloading path during cassava enlargement. Using real-time fluorescence quantitative PCR technique, △△ Ct method was used to determine relative expression of sucrose metabolism enzyme m RNA. The regulation of extracellular body pathway Both the enzyme cell wall and soluble acid invertase showed higher expression in the fibrous roots, especially the expression of cell wall acidic invertase in both fibrous roots was significantly higher than that in three developmental stages On the contrary, the expression level of sucrose synthase, which is the regulation enzyme of the plastid pathway, was significantly higher in tuberous roots than that in fibrous roots. In particular, the expansive stage exhibited the highest level of expression in these tissues. Therefore, the relatively high expression of cell wall acid invertase and sucrose synthase in the fibrous roots and tuberous roots, respectively, preliminarily confirmed the dominant position of the plastidal unloading pathways of the apoplast and root of the fibrous roots.
