目的 研究幽门螺杆菌 (H .pylori)感染对胃癌及癌旁组织中Bcl 2家族基因表达的影响。方法 收集 95例胃癌及癌旁组织标本 ,应用半定量RT PCR法检测Bid、Bax和Bcl 2mRNA的表达。结果 胃癌组织中Bid和BaxmRNA表达按H .pylori阴性组、Ca gA-H .pylori感染组和CagA+ H .pylori感染组顺序依次增高 (P <0 0 5 ) ,H .pylori阴性组的Bcl 2mRNA表达低于H .pylori阳性组 (P<0 0 5 ) ;癌旁组织中 ,Bid、Bax和Bcl 2表达按H .pylori阴性组、CagA-H .pylori感染组和CagA+ H .pylori感染组顺序均依次递增 (P <0 0 5 )。H .pylori阴性组中 ,胃癌组织的Bid和Bax表达与Bcl 2表达呈负相关 (P <0 0 5 ) ,而在H .pylori感染组中 ,胃癌组织的Bid和Bax表达与Bcl 2表达无相关性 ;癌旁组织的Bid和Bax表达与Bcl 2表达呈正相关 (P <0 0 5 )。结论 H .pylori有促进Bid、Bax和Bcl 2表达的作用 ,并且能够诱发这些细胞凋亡调控基因的表达紊乱 ,这也许是H .pylori诱导胃癌发生的途径之一。
Objective To investigate the effect of Helicobacter pylori infection on the gene expression of Bcl-2 in gastric cancer and its adjacent tissues. Methods Ninety-five gastric and para-cancerous tissues were collected and the expression of Bid, Bax and Bcl-2 were detected by semi-quantitative RT-PCR. Results The expression of Bid and Bax mRNA in gastric cancer tissues was increased in order of H.pylori negative group, Ca gA-Hpylori infection group and CagA + H.pylori infection group (P <0 05), and Bcl 2 mRNA expression in H.pylori negative group (P <0.05). The expression of Bid, Bax and Bcl2 in the adjacent tissues of H. pylori were lower than those in the H.pylori negative group, CagA-Hpylori infection group and CagA + H.pylori infection group Followed by increasing (P <0 05). In H.pylori-negative group, the expression of Bid and Bax in gastric cancer tissues was negatively correlated with the expression of Bcl-2 (P <0.05), while in H.pylori infection group, the expression of Bid and Bax in gastric cancer tissues was negatively correlated with the expression of Bcl-2 The expression of Bid and Bax in paracancerous tissues was positively correlated with the expression of Bcl 2 (P <0.05). Conclusions Hpylori can promote the expression of Bid, Bax and Bcl2, and induce the expression disorder of these apoptosis-controlling genes, which may be one of the pathways by which H. pylori induces gastric cancer.
