本文报道了草菇(Volvariella volvacea)和银丝草菇(V.bombycina)菌丝原生质体的分离、纯化、培养和再生。结果表明:1.溶壁酶、纤维素酶能破壁分离原生质体,β-葡糖苷酸酶有增效作用;2.用于分离原生质体的菌丝年龄以2天为宜,酶解时间2-4小时即可;3.所得的原生质体活力可达90.9-91.8%;4.原生质体再生菌丝有多种方式。
This paper reports the isolation, purification, cultivation and regeneration of mycelium protoplasts of Volvariella volvacea and V.bombycina. The results showed that: 1. Lysozyme and cellulase could break the protoplasts and the synergism of β-glucuronidase could be broken; 2. The suitable hyphae age for separating protoplasts was 2 days, 2-4 hours; 3. The resulting protoplast activity up to 90.9-91.8%; 4. Protoplast regenerated mycelium in a variety of ways.
