目的观察不同浓度四君子汤对尾部悬吊大鼠比目鱼肌肌萎缩的影响。方法尾部悬吊法建立模拟失重模型,30只Sprague-Dawley大鼠随机分为5组,同步饲养组(CON)、尾吊组(HLU-W)、尾吊+四君子汤高(HLU-H)、中(HLU-M)、低(HLU-L)浓度组,每组6只。用免疫组织化学法观察药物对比目鱼肌肌球蛋白重链(MHC)的影响。结果与尾吊组相比,四君子汤高、中浓度组使比目鱼肌湿重分别增加了33.11%和37.57%,与同步饲养组亦无显著性差异;四君子汤高、中、低浓度组使比目鱼肌Ⅰ型肌纤维比例增加了34.04%~54.90%,Ⅰ型肌横截面积增加了8.50%~36.86%,且中浓度组与同步饲养组相比亦无显著性差异。结论3种浓度的四君子汤对模拟失重引起的比目鱼肌肌萎缩有显著的对抗作用,且中浓度四君子汤的效果优于高、低浓度。
Objective To observe the effects of different concentrations of Sijunzi Decoction on muscle atrophy of soleus in tail-suspended rats. Methods 30 rats were randomly divided into 5 groups: CON, HLU-W, HLU-H, (HLU-M), low (HLU-L) concentration group, 6 in each group. The effects of the drugs on the myosin heavy chain (MHC) of the soleus muscle were observed by immunohistochemistry. Results Sijunzi decoction increased the soleus muscle wet weight by 33.11% and 37.57%, respectively, compared with the tail suspension group, and also had no significant difference with the synchronous rearing group. The Sijunzi decoction high, middle and low concentration groups increased the soleus Muscle type Ⅰ muscle fibers increased by 34.04% to 54.90%, type Ⅰ muscle increased by 8.50% to 36.86%, and the concentration group and synchronous feeding group compared with no significant difference. Conclusion Sijunzi Decoction with three concentrations has significant antagonistic effect on soleus muscle atrophy caused by simulated weightlessness, and the effect of medium concentration Sijunzi Decoction is better than that of high and low concentration.
