目的基于基因克隆对远志鲨烯环氧酶(SE)进行蛋白结构及进化分析。方法克隆远志鲨烯环氧酶基因(SE)DNA片段,将已克隆SE基因翻译成氨基酸序列,进行蛋白结构及进化分析。结果已克隆远志鲨烯环氧酶基因DNA片段长517bp,具有170个氨基酸,有三个特征位点:Protein kinase C phosphorylation site、N-myristoylation site与Tyrosine kinase phosphorylation site,远志SE基因与Medicago sativa和Medicago truncatula的SE基因亲缘关系较为接近。结论已克隆远志SE基因DNA片段能够推测远志鲨烯环氧酶部分特征位点,可以进行远志SE蛋白进化分析。
OBJECTIVE: To analyze the protein structure and evolution of aspartic acid epoxidase (SE) based on gene cloning. Methods The fragment of asparagine epoxidase gene (SE) was cloned. The cloned SE gene was translated into the amino acid sequence for protein structure and evolution analysis. Results The DNA fragment of asparagine epoxidase gene was cloned and its length was 517 bp with 170 amino acids. The DNA fragment contained three characteristic sites: the protein kinase C phosphorylation site, the N-myristoylation site and the Tyrosine kinase phosphorylation site. The Polygala SE gene was associated with Medicago sativa and Medicago SE gene of truncatula is more closely related. Conclusion The DNA fragment of Polygala tenuifaciens SE gene has been cloned to speculate part of polygalacturonase epoxidase characteristic sites, the polygalaceae SE protein evolution analysis.
