目的观察溃疡平对胃溃疡大鼠胃黏膜组织血管内皮生长因子(VEGF)和血管内皮生长因子受体-1(Flt-1)mRNA表达的影响,探讨溃疡平促进大鼠胃溃疡愈合的机制。方法采用改良Okabe法制备慢性乙酸胃溃疡模型,随机分为:正常对照组,模型对照组,溃疡平高、中、低剂量(5.6、2.8、1.4g/kg)组,盐酸雷尼替丁(28.3mg/kg)组。给药治疗14d,采用实时荧光定量PCR法检测胃黏膜组织VEGF和Flt-1mRNA表达。结果与模型对照组相比,溃疡平中、高剂量组的VEGF2-ΔΔCT值明显增高,表明溃疡平中、高剂量组的VEGFmRNA表达上调,有统计学意义(P分别<0.05和<0.001);溃疡平高剂量组的Flt-12-ΔΔCT值明显增高,表明溃疡平高剂量组的Flt-1mRNA表达上调,有统计学意义(P<0.001)。结论溃疡平可促进胃溃疡大鼠胃黏膜组织VEGF和Flt-1mRNA表达,这可能是其促进溃疡愈合,提高溃疡愈合质量,减少溃疡复发的分子学机制之一。
Objective To observe the effect of ulcer on the expression of vascular endothelial growth factor (VEGF) and vascular endothelial growth factor receptor-1 (Flt-1) mRNA in gastric mucosa of rats with gastric ulcer, and to explore the mechanism of ulcer in promoting the healing of gastric ulcer in rats. Methods The modified Okabe method was used to prepare chronic acetic acid gastric ulcer model and randomly divided into normal control group, model control group, flat ulcer, moderate and low dose (5.6,2.8 and 1.4g / kg), ranitidine hydrochloride 28.3 mg / kg) group. After 14 days of treatment, the expression of VEGF and Flt-1mRNA in gastric mucosa were detected by real-time fluorescence quantitative PCR. Results Compared with the model control group, the VEGF2-ΔΔCT value of the middle and high dose ulcer group was significantly higher than that of the model control group, indicating that the expression of VEGFmRNA in the medium and high dose ulcer groups was up-regulated with statistical significance (P <0.05 and <0.001, respectively) The level of Flt-12-ΔΔCT in the ulcer-treated group was significantly higher than that in the untreated group, indicating that the level of Flt-1 mRNA in the ulcer-treated high dose group was significantly increased (P <0.001). Conclusion Ulcer can promote the expression of VEGF and Flt-1mRNA in gastric mucosa of gastric ulcer rats, which may be one of the molecular mechanisms that promote the healing of ulcer, improve the healing quality of ulcer and reduce the recurrence of ulcer.
