目的:优选白纸扇感冒颗粒中挥发油β-环糊精(β-CD)包合物的制备工艺。方法:以挥发油包合率、包合物收得率及包合物含油率为指标,从饱和水溶液法、超声法、研磨法中挑选最佳制备方法;以挥发油包合率和包合物收得率为综合评价指标,选取挥发油与β-CD投料比、包合温度、包合时间、β-CD与水的比例为影响因素,采用正交试验优选挥发油包合工艺;应用TLC,UV及显微成像对制备的包合物进行物相鉴别。结果:最佳的包合方法为饱和水溶液法,其最佳包合工艺为挥发油-β-CD 1∶8,包合温度55℃,搅拌时间3 h,β-CD-水1∶10;TLC,UV及显微成像分析结果均显示挥发油与β-CD已形成包合物。结论:优选的包合工艺简单、稳定可行,适合工业化大生产。
Objective: To optimize the preparation process of volatile oil β-cyclodextrin (β-CD) inclusion complex in Baizhang Fan. Methods: The optimum preparation method was selected from the methods of saturated aqueous solution, ultrasonic method and grinding method with the inclusion rate of volatile oil, the yield of inclusion compound and the oil content of inclusion compound. The yield was a comprehensive evaluation index. The ratio of β-CD to volatile oil, inclusion temperature, inclusion time and the ratio of β-CD to water were chosen as influencing factors. Orthogonal test was adopted to optimize the inclusion process of volatile oil. Microscopic imaging of the inclusion complex phase identification. Results: The best inclusion method was saturated aqueous solution method, the best inclusion process was volatile oil-β-CD 1:8, inclusion temperature 55 ℃, stirring time 3 h, β-CD-water 1:10; TLC The results of UV and microscopic imaging showed that the essential oil and β-CD had formed inclusion complex. Conclusion: The preferred inclusion process is simple, stable and feasible, suitable for large-scale industrial production.
