目的:探讨P物质(substance P,SP)对过敏小鼠腹腔灌洗液中巨噬细胞表面蛋白酶激活受体(protease activated recep-tors,PARs)表达的影响。方法:通过腹腔注射鸡卵清蛋白(ovalbumin,OVA)建立过敏小鼠模型,再根据腹腔注射的SP的浓度将小鼠分为生理盐水组、0.2、2.0、20.0和200.0μg/ml SP组,每组6只,同时每组均设有未致敏的对照组。最后一次激发3 h后处死小鼠,取腹腔灌洗液细胞经固定,用流式细胞仪检测巨噬细胞表面PARs的表达。结果:过敏组小鼠腹腔灌洗液中巨噬细胞表面PARs的表达水平较正常组明显降低(P<0.01),而不同浓度的SP组之间无统计学差异(P>0.05)。结论:SP对小鼠腹腔巨噬细胞PARs表达无影响,但过敏原可影响巨噬细胞表面PARs的表达。
Objective: To investigate the effect of substance P (SP) on the expression of proteasome-activated recep-tors (PARs) in macrophages in peritoneal lavage fluid of allergic mice. Methods: The allergic mouse model was established by intraperitoneal injection of ovalbumin (OVA). According to the concentration of SP injected intraperitoneally, the mice were divided into normal saline group, 0.2, 2.0, 20.0 and 200.0 μg / ml SP groups, 6 in each group, while each group was provided with a non-sensitized control group. The mice were sacrificed 3 h after the last challenge. The peritoneal lavage fluid was fixed and the expression of PARs on macrophages was detected by flow cytometry. Results: The expression of PARs on macrophages in peritoneal lavage fluid of allergic mice was significantly lower than that of normal mice (P <0.01), but there was no significant difference among different concentrations of SP (P> 0.05). CONCLUSIONS: SP has no effect on PARs expression in mouse peritoneal macrophages, but allergens can affect the expression of PARs on macrophages.
