目的探讨骨灵膏及其拆方制剂对OA大鼠T淋巴细胞CD3+、CD4+、CD8+及CD80、CD86、COL-Ⅱ与OA的作用机制。方法 60只雌性SD大鼠随机分为GLG、GUG、LG、CLXB、M及NG等6组,每组10只,采用冷固法建立OA模型并给予GLG等灌胃10周,取大鼠血浆、膝关节滑膜及软骨组织;采用免疫荧光检测血浆及滑膜CD3+、CD4+、CD8+的阳性细胞率,流式细胞术检测软骨CD3+、CD4+、CD8+及CD80、CD86、Col-Ⅱ百分率。结果 M组与NG组比较能降低血浆及软骨CD3+、CD4+T淋巴细胞百分率(P<0.01),增高滑膜CD3+、CD4+、CD8+、CD4+/CD8+百分率和软骨CD80、CD86、COL-Ⅱ阳性百分率(P<0.01);GLG与M组比较能升高血浆及软骨CD3+、CD4+百分率(P<0.01),降低软骨CD80、CD86、COL-Ⅱ阳性百分率和滑膜CD4+/CD8+比值(P<0.01);GLG与GUG、LG、CLXB组比较能升高血浆及软骨CD3+、CD4+百分率及CD4+/CD8+比值(P<0.01)。结论 GLG可能通过提高OA大鼠整体和局部关节免疫功能,抑制软骨细胞APC功能的激活和抗自身抗原的免疫反应,减轻了滑膜免疫炎症反应,保护OA软骨和滑膜,作用优于其拆方制剂及CLXB。
OBJECTIVE To investigate the mechanism of Bailinggao and its disassembled preparations on the T lymphocyte CD3 +, CD4 +, CD8 +, CD80, CD86 and COL-Ⅱ in OA rats. Methods Sixty female Sprague-Dawley rats were randomly divided into six groups (GLG, GUG, LG, CLXB, M and NG), with 10 rats in each group. OA models were established by cold- , Synovium and cartilage in knee joint. The positive rate of CD3 +, CD4 + and CD8 + in plasma and synovium was detected by immunofluorescence. The percentage of CD3 +, CD4 +, CD8 +, CD80, CD86 and Col-Ⅱ in cartilage were detected by flow cytometry. Results The percentage of CD3 +, CD4 +, CD8 +, CD4 + / CD8 + and CD80, CD86 and COL-Ⅱ in plasma and cartilage were decreased in M and NG groups (P <0.01) (P <0.01). Compared with M group, GLG increased the percentage of CD3 + and CD4 + in plasma and cartilage (P <0.01) and decreased the positive percentage of CD80, CD86 and COL-Ⅱ in cartilage and the ratio of CD4 + / CD8 + ; Compared with GUG, LG and CLXB groups, GLG could increase the ratio of CD3 +, CD4 + and CD4 + / CD8 + in plasma and cartilage (P <0.01). Conclusion GLG can relieve the synovial immune-inflammation reaction and protect OA cartilage and synovium, which is better than its disassembly by improving the global and regional joint immune function, inhibiting the activation of APC function and the anti-autoantigen immune response of chondrocytes.
