目的:观察“标本配穴”电针对慢性心肌缺血大鼠缺血心肌缝隙连接蛋白43(Cx43)表达的影响,探讨“标本配穴”电针防治心肌缺血的作用机制。方法:采用SPF级雄性Wistar大鼠60只,随机分为4组:正常组、模型组、内关治疗组、标本配穴治疗组,每组15只。除正常组连续14d皮下注射0.9%氯化钠溶液外(2mg·kg~(-1)·d~(-1)),其他3组均每日给予盐酸异丙肾上腺素(2mg·kg~(-1)·d~(-1))皮下注射,每日1次,连续14d。两个治疗组在造模开始当天即给予电针针刺治疗,正常组和模型组相同时间抓取固定,每日1次,共21d。采用ELISA检测各组大鼠MDA、SOD、VCAM-1,Western blot及免疫组化法检测Cx43表达及分布。结果:与正常组比较,模型组MDA、VCAM-1含量升高,总SOD活力、心肌Cx43蛋白表达降低(P<0.05);与模型组比较,内关治疗组、标本配穴组MDA、VCAM-1含量降低,总SOD活力、心肌Cx43蛋白表达升高(P<0.05);标本配穴组总SOD活力、VCAM-1含量均低于内关治疗组,Cx43蛋白表达高于内关治疗组(P<0.05)。结论:电针可通过促进Cx43的表达,改善其分布,保护或者修复缝隙连接使其发挥保护心肌的作用;“标本配穴”电针可更好的通过促进Cx43的表达和改善心肌Cx43的分布,起到保护慢性心肌缺血模型大鼠心肌的作用。
Objective: To observe the effect of “acupoints of acupoints” electroacupuncture on the expression of connexin 43 (Cx43) in ischemic myocardium in rats with chronic myocardial ischemia, and to explore the mechanism of “acupoints of acupoints” and electroacupuncture on the prevention and treatment of myocardial ischemia. Methods: Sixty SPF male Wistar rats were randomly divided into 4 groups: normal group, model group, Neiguan treatment group and specimen with acupoint treatment group, 15 in each group. The rats in the other three groups were given isoproterenol hydrochloride (2 mg · kg ~ (-1) · d ~ (-1) · d ~ (-1) -1) · d ~ (-1)) subcutaneously once daily for 14 days. The two treatment groups were given electroacupuncture acupuncture treatment on the day of modeling, the normal group and the model group were fixed at the same time, once a day for 21 days. The expressions of MDA, SOD, VCAM-1, Western blot and immunohistochemistry were used to detect the expression and distribution of Cx43 in each group. Results: Compared with normal group, the content of MDA and VCAM-1 in model group increased, while the activity of total SOD and the expression of Cx43 protein in myocardium decreased (P <0.05). Compared with model group, (P <0.05). The total SOD activity and the level of VCAM-1 in the acupoints group were lower than those in the Neiguan group and the expression of Cx43 protein was higher than that in the Neiguan group (P <0.05). Conclusion: Electroacupuncture can improve the distribution of Cx43, protect or repair the gap junction and play a role in protecting the myocardium. Electroacupuncture can improve the expression of Cx43 and improve the expression of Cx43 Of the distribution, play a role in protecting the myocardium of chronic myocardial ischemia model rats.
