目的观察大黄蟅虫丸对胰岛素样生长因子(IGF-1)在动脉粥样硬化(AS)模型大鼠中表达的影响,并从中探讨大黄蟅虫丸抗AS的机制。方法SD雄性大鼠体重220~300g,随机分为4组,对照组、模型组、大黄蟅虫丸组高剂量、低剂量组,每组8只,共32只。用高脂饲料喂养并建立大鼠AS模型,用大黄蟅虫丸灌胃12w后,观察各组大鼠血清血脂(TG,TC,HDL-C,LDL-C),丙二醛(MDA),超氧化物歧化酶(SOD),IGF-1含量的变化。结果模型组IGF-1含量显著降低,与对照组比较有显著差异(P<0.05);大黄蟅虫丸组较模型组血清IGF-1含量明显升高(P<0.05)。结论大黄蟅虫丸通过提高血清IGF-1表达,抑制血管平滑肌细胞的凋亡,发挥抗AS的作用。
Objective To observe the effect of Dahuangqichong Pill on the expression of insulin-like growth factor (IGF-1) in rats with atherosclerosis (AS), and to discuss the mechanism of anti-AS of Dahuangqichong Pill. Methods SD male rats weighing 220-300g were randomly divided into 4 groups: control group, model group, and high-dose and low-dose group of Dahuangqichong Pills group, with 8 rats in each group, a total of 32 animals. Rats were fed with a high-fat diet and established a rat model of AS. After a 12-hour gavage of Dahuangqichong Pill, serum lipids (TG, TC, HDL-C, LDL-C) and malondialdehyde (MDA) were observed in each group. Changes of superoxide dismutase (SOD) and IGF-1 content. Results The content of IGF-1 in the model group was significantly lower than that in the control group (P<0.05). The level of serum IGF-1 in the Dahuangzhiceng Pill group was significantly higher than that in the model group (P<0.05). Conclusion Dahuangqichong Pill plays an anti-AS role by increasing serum IGF-1 expression and inhibiting apoptosis of vascular smooth muscle cells.
