目的建立稳定持久的大鼠慢性高眼压模型,为青光眼的各项研究奠定基础。方法雌性SD大鼠20只,随机分为单纯手术组10只,单纯烙闭右眼3支上巩膜静脉;手术加药物组10只,烙闭右眼3支上巩膜静脉合并烙闭处结膜下放置丝裂霉素C棉片;两组左眼均为对照眼。两组于术后分别测术后即刻、术后1h、1d、1、2、4、8周的眼压,并观察两组眼术后局部表现。结果单纯手术组于即刻、术后1h、1d、1、2周眼压均高于对照组(P<0.05),术后4周眼压(9.85±1.06)mmHg,8周(9.31±0.85)mmHg,与对照组比较差异无统计学意义(P>0.05);手术加药物组于术后即刻、术后1h、1d、1、2、4、8周的眼压均显著高于对照组(P<0.05)。结论用烙闭上巩膜静脉合并丝裂霉素C棉片贴敷方法制作的慢性高眼压动物模型操作简单,高眼压维持时间较长,成模率高,为较理想的慢性高眼压模型。
Objective To establish a stable and long-term model of chronic ocular hypertension in rats and lay the foundation for the various studies of glaucoma. Methods Twenty female Sprague-Dawley rats were randomly divided into three groups: the simple operation group (3 eyes), the left scleral vein alone (3 eyes), and the surgery plus drug group (10 eyes) Place mitomycin C cotton; both left eye are control eyes. The intraocular pressure of the two groups immediately after operation, 1h, 1d, 1, 2, 4 and 8 weeks after operation were observed, and the local postoperative performance was observed. Results The intraocular pressure (IOP) at 1h, 1d, 1 and 2 weeks postoperatively in the surgery group was significantly higher than that in the control group (9.85 ± 1.06 mmHg, 9.31 ± 0.85, mmHg, there was no significant difference compared with the control group (P> 0.05). The intraocular pressure of the operation plus drug group immediately after operation, 1h, 1d, 1, 2, 4 and 8 weeks after operation was significantly higher than that of the control group P <0.05). Conclusion The model of chronic ocular hypertension with branching and closing of the scleral vein combined with mitomycin C cotton patch has the advantages of simple operation, long maintenance of intraocular pressure and high rate of mold formation, which is an ideal chronic ocular hypertension model.
