目的:研究复方丹参滴丸滴制料液中丹参素、原儿茶醛含量的快速分析方法,及化料过程的均一性在线监测技术。方法:以高效液相色谱分析值为参照,创新采用近红外透反射光谱技术采集料液NIR光谱,对光谱进行波长选择和预处理,采用偏最小二乘回归建立校正模型,并使用建立好的PLSR定量模型对料液的丹参素、原儿茶醛含量进行快速检测,对化料过程均一性进行在线监测,确定化料终点。结果:模型性能良好,丹参素、原儿茶醛的相关系数分别为0.9929和0.9937,验证集均方差分别为0.368和0.159。NIR光谱法可实时反映料液中采集点位置的成分含量变化,并正确反映化料过程中料液均一状态。结论:近红外透反射光谱技术可有效采集滴丸料液NIR光谱,近红外光谱法可快速准确测定滴丸料液中成分含量,可用于滴丸剂料液均一性的判断。
Objective: To study the rapid analysis method of Danshensu and protocatechuic aldehyde content in compound Danshen dropping pills and its homogeneity on-line monitoring technology. Methods: Based on the analytical value of high performance liquid chromatography (HPLC), the NIR spectra of liquid samples were collected using NIR transmission spectroscopy. The wavelength of the spectrum was selected and pretreated. The calibration model was established by partial least squares regression. PLSR quantitative model of the liquid danshensu, protocatechuic aldehyde content for rapid detection of the homogeneity of the chemical process on-line monitoring to determine the end of the chemical material. Results: The performance of the model was good. The correlation coefficients of Danshensu and protocatechuic aldehyde were 0.9929 and 0.9937, respectively. The mean square error of validation was 0.368 and 0.159 respectively. NIR spectroscopy can reflect the change of the component content of the collection point in the feed liquid in real time and correctly reflect the uniform state of the feed solution in the feedstock. Conclusion: Near Infrared Transmittance Spectroscopy (NIRS) can effectively collect the NIR spectrum of drip liquid and NIR spectroscopy can quickly and accurately determine the content of drip liquid. It can be used to judge the homogeneity of drip liquid.
