目的:用毛细管气相色谱法测定海狗油中二十碳五烯酸(EPA)、二十二碳五烯酸(DPA)和二十二碳六烯酸(DHA)的含量。方法:采用HP-INNOWAX毛细管柱(30 m×0.320 mm,0.25 μm),柱温150℃,保持5 min后,以10℃·min-1升至200℃后,继续以5℃·min-1升至240℃,保持7 min,载气为氮气(99.999%)。进样口温度为240℃,检测器温度为270℃,FID检测器。结果:EPA甲酯平均回收率为99.5%,RSD为1.2%;DPA甲酯平均回收率为98.8%,RSD为1.1%;DHA甲酯平均回收率为98.5%,RSD为0.9%。结论:此法操作简单、准确,重复性、稳定性好,是一种可行的ω-3多不饱和脂肪酸含量测定方法。
Objective: To determine the content of eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA) and docosahexaenoic acid (DHA) in seal oil by capillary gas chromatography. METHODS: HP-INNOWAX capillary column (30 m × 0.320 mm, 0.25 μm) was used. The column temperature was maintained at 150 ℃ for 5 min and then increased to 200 ℃ at 10 ℃ · min-1. The temperature was raised to 240 ° C for 7 min and the carrier gas was nitrogen (99.999%). Inlet temperature is 240 ° C, detector temperature is 270 ° C, FID detector. Results: The average recovery rate of EPA methyl ester was 99.5% and the RSD was 1.2%. The average recovery of methylparaben was 98.8% and RSD was 1.1%. The average recovery of methyl ester of DHA was 98.5% and the RSD was 0.9%. Conclusion: This method is simple, accurate, reproducible and stable. It is a feasible method for the determination of ω-3 polyunsaturated fatty acids.
