目的:建立胶原诱导性关节炎(C IA)大鼠模型,观察姜黄属常用中药姜黄、郁金、莪术对C IA大鼠滑膜炎症的影响,比较它们的抗炎作用差异。方法:将50只SD大鼠随机分为正常对照组、模型组、姜黄组、郁金组、莪术组。除正常对照组同法注射等量生理盐水外,余各组采用右足趾部皮内注射胶原乳液免疫建立C IA模型。7天后,大鼠尾根部皮内注射胶原乳化液加强免疫1次。初次免疫21天后开始分组给药,姜黄组、郁金组及莪术组分别以中药姜黄、郁金、莪术100%水煎液灌胃;正常对照组、模型组以等量生理盐水灌胃,连续14天。造模后观察各组大鼠的一般情况、关节肿胀程度,计算肿胀率。给药14天后取大鼠左膝关节滑膜组织,固定、包埋、切片、HE染色,观察各组大鼠滑膜组织病理学特征。结果:除正常对照组外,余各组大鼠左足关节明显肿胀,关节肿胀率与正常组比较有显著性差异(P<0.01),说明造模成功;给药14天后,姜黄组、莪术组与模型组比较踝关节肿胀程度减轻,有统计学意义(P<0.05)。病理观察显示,姜黄组、郁金组、莪术组较模型组大鼠滑膜组织充血减轻,炎性细胞浸润减少,以姜黄组最为明显。结论:姜黄属常用中药均能减轻C IA大鼠滑膜炎症反应,其中,以姜黄抗关节炎作用最强。
OBJECTIVE: To establish a rat model of collagen-induced arthritis (C IA) and observe the effect of curcumin, turmeric and curcuma on the inflammation of synovium of CIA rats, and compare their anti-inflammatory effects. Methods: Fifty SD rats were randomly divided into normal control group, model group, turmeric group, turmeric group and zedoary group. Except for the normal control group, the same amount of saline was injected in the same way, while the C IA model was established by intradermal injection of collagen emulsion in the right toe of each group. Seven days later, the rat tail root intradermal injection of collagen emulsion to enhance immunity 1. After 21 days of initial immunization, the rats were given the drug in groups. The turmeric group, the turmeric group and the Curcuma group were respectively gavaged with turmeric, turmeric and curcuma 100% water decoction. The normal control group and the model group were given gavage with normal saline, 14 days. After modeling, the general situation of rats in each group was observed, the extent of joint swelling and the swelling rate were calculated. After 14 days of administration, the left knee joint synovial tissue of rats was taken, fixed, embedded, sectioned and stained with HE. The pathological features of synovial tissue in each group were observed. Results: In addition to the normal control group, the left foot joints in each group were significantly swollen and the swollen joint rate was significantly different from that in the normal group (P <0.01), indicating successful modeling. After 14 days of administration, Compared with the model group, the degree of ankle swelling was reduced, with statistical significance (P <0.05). Pathological observation showed that the turmeric group, the turmeric group, the Curcuma group than the model group rats synovial tissue congestion, inflammatory cell infiltration decreased, the most obvious turmeric group. Conclusion: Curcumin commonly used traditional Chinese medicine can reduce C IA rat synovial inflammatory response, of which the role of the strongest anti-arthritis turmeric.
