目的 :研究雄激素对脑穿刺损伤后星形胶质细胞(Ast)反应性胶质化的影响。方法 :通过免疫组化、免疫荧光染色及流式细胞仪计数等方法 ,观察切除睾丸及补充睾丸酮后脑穿刺损伤鼠伤灶周围GFAP IR阳性细胞的形态、数量与比例变化。结果 :切除睾丸鼠脑损伤后 ,伤灶周围反应性胶质比较单纯损伤鼠明显 ,表现为胞体肥大 ,突起增粗、延长 ,GFAP免疫组化染色显著增强。睾丸酮治疗后 ,Ast反应程度虽有减弱 ,但未能恢复到正常水平。流式细胞计数结果表明 ,切除睾丸及用睾丸酮治疗前后 ,Ast比例无显著变化。结论 :睾丸酮能影响Ast的形态变化及对损伤的反应 ,但可能对Ast的增殖作用不明显。
Objective: To investigate the effect of androgens on reactive gliosis of astrocytes after brain puncture injury. Methods: Immunohistochemistry, immunofluorescence staining and flow cytometry were used to observe the morphological, quantitative and proportion changes of GFAP-IR positive cells around the lesion of the lesion after the excision of the testis and the supplementation of testosterone. Results: After excision of testicular brain injury, the reactive glia around the lesion was more obvious than pure injury, which was characterized by hypertrophy of the cell body, thickening and prolongation of the protuberance, and significant enhancement of GFAP immunohistochemical staining. After testosterone treatment, Ast response although weakened, but failed to return to normal levels. Flow cytometry results showed that before and after excision of testis and testosterone treatment, there was no significant change in the proportion of Ast. Conclusion: Testosterone can affect the morphological changes of Ast and its response to injury, but may not have a significant effect on the proliferation of Ast.
