目的 比较粘附细胞作为APC能否较一般方法提高T细胞对神经髓鞘类抗原的增殖反应性和显示出MS对神经髓鞘类抗原的特异自身免疫反应特征。方法 选HLA DR15亚型MS和正常人外周血单个核细胞 ,体外培养。分别用一般T淋巴细胞增殖试验和富集粘附细胞法检测对人神经髓鞘、蛋白脂蛋白 (PLP)和碱性髓鞘蛋白(MBP)淋巴细胞增殖反应性。结果 富集粘附细胞法使MS组和对照组T淋巴细胞对神经髓鞘、PLP和MBP的增殖较标准法提高 (P <0 .0 5 ) ;使MS组以MBP和PLP的增殖反应较对照组的差别具有显著性的意义 ;而对TT无明显提高作用。结论 富集粘附细胞法可提高T淋巴细胞对神经髓鞘、PLP和MBP的增殖反应性 (MS和NC) ,多数情况使MS组和NC组的差别具有显著性的意义 ,对非髓鞘类抗原 (如TT)无这种作用。
OBJECTIVE: To compare whether adherent cells act as APCs to enhance the proliferative reactivity of T cells to myelin antigens and to reveal the specific autoimmune responses of MS to neuromuscular antigens. Methods HLA DR15 subtype MS and normal human peripheral blood mononuclear cells were cultured in vitro. The proliferative reactivity to human myelin, proteolipoprotein (PLP) and basic myelin protein (MBP) lymphocytes were detected by T-lymphocyte proliferation assay and cell adhesion-enrichment assay respectively. Results Enrichment of adherent cells increased the proliferation of myelin sheath, PLP and MBP in MS group and control group compared with the standard method (P <0.05). Compared with normal group, the proliferative response of MS group and control group to MBP and PLP The difference between the control group has a significant significance; while no significant effect on TT. Conclusion Enrichment of adherent cells can increase the proliferative reactivity (MS and NC) of T lymphocytes to myelin sheath, PLP and MBP. In most cases, the difference between MS group and NC group is significant, Antigens such as TT do not have this effect.
