目的探讨白细胞介素6(IL-6)与活化诱导胞嘧啶核苷脱氨酶(AICD)在结直肠癌前病变表达的关系及消痰通腑方的干预作用。方法 AOM/DSS联合造模法复制小鼠结直肠癌前病变模型,随机分为空白组、模型组、中药组、西药组、中西医结合组。空白组、模型组给予等量蒸馏水;中药组给予消痰通腑方(26 g·kg-1·d-1)浓缩液0.2 ml/d;西药组每只小鼠给予柳氮磺吡啶(7.8 mg/d);中西医结合组同时给予消痰通腑方和柳氮磺吡啶。各组均连续给药12周,HE染色观察小鼠肠黏膜上皮组织形态及不典型增生,免疫组化及酶联免疫吸附测定法(ELISA)检测IL-6、AICD表达情况。结果与正常组比较,模型组IL-6和AICD免疫组化指数、血清蛋白表达升高,差异有统计学意义(P<0.01)。与模型组比较,18周中药组、西药组、中西医结合组IL-6和AICD免疫组化指数、血清蛋白表达下降,差异有统计学意义(P<0.05)。与西药组比较,18周中药组AICD的免疫组化指数表达下降,差异有统计学意义(P<0.05)。结论消痰通腑方能够降低肠道内炎症因子IL-6及AICD的表达,从而降低发生体细胞高频突变(SHM)和类别转换重组(CSR)的机率,达到防治结直肠癌前病变的作用。
Objective To investigate the relationship between interleukin-6 (IL-6) and activation-induced cytidine deaminase (AICD) expression in colorectal precancerous lesions and the therapeutic effect of Xiaotan Tongfu recipe. Methods A mouse model of colorectal precancerous lesions was duplicated by AOM / DSS combined with modeling and randomly divided into blank group, model group, traditional Chinese medicine group, western medicine group and traditional Chinese and western medicine combination group. The rats in the blank group and the model group were given the same amount of distilled water. The Chinese medicine group was given 0.2 ml / d of the concentrate of Xiaotan Tongfu Formula (26 g · kg -1 · d -1) and the sulfasalazine mg / d). The combination of traditional Chinese and western medicine was given Xiaotan Tongfu prescription and sulfasalazine. The rats in each group were given continuous administration for 12 weeks. The morphology and atypical hyperplasia of intestinal mucosa in mice were observed by HE staining. The expression of IL-6 and AICD was detected by immunohistochemistry and enzyme-linked immunosorbent assay (ELISA). Results Compared with the normal group, the immunohistochemical indexes of IL-6 and AICD and the expression of serum protein in the model group were significantly increased (P <0.01). Compared with the model group, the immunohistochemical indexes of IL-6 and AICD in 18-week Chinese medicine group, Western medicine group and Integrative Chinese and Western medicine groups decreased, the serum protein expression decreased, the difference was statistically significant (P <0.05). Compared with Western medicine group, the expression of AICD in 18-week Chinese medicine group decreased significantly, with statistical significance (P <0.05). Conclusion Xiaotan Tongfu Decoction can reduce the expression of inflammatory factors IL-6 and AICD in the intestine and thus reduce the incidence of somatic high-frequency mutation (SHM) and type-transfer recombination (CSR) and achieve the prevention and treatment of colorectal precancerous lesions .
