目的观察重组Galectin-9腺病毒对小鼠心脏移植物生存期及调节性T(Treg)细胞分化和增殖的影响。方法建立小鼠心脏移植模型,随机均分为3组(n=10):对照组(术后用0.9%NaCl溶液)、空腺病毒组、重组Galectin-9腺病毒组,记录心脏移植物生存时间,于移植心停搏或术后14d取脾,分离单个核细胞,流式细胞仪检测CD4+CD25+Treg细胞比例。ELISA法检测外周血IL-10、TGF-β1浓度。结果与对照组、空腺病毒组相比,重组Galectin-9腺病毒组心脏移植物生存时间明显升高(P<0.01),重组Galectin-9腺病毒组CD4+CD25+Treg细胞比例明显升高(P<0.01)。重组Galectin-9腺病毒组IL-10、TGF-β1的含量明显高于对照组及空腺病毒组(P<0.01)。结论重组Galectin-9腺病毒能促进体内CD4+CD25+Treg细胞的分化与增殖,明显提高小鼠心脏移植物的生存时间。
Objective To observe the effect of recombinant Galectin-9 adenovirus on cardiac allograft survival and regulatory T (Treg) cell differentiation and proliferation in mice. Methods The mouse heart transplantation model was established and randomly divided into three groups (n = 10): control group (0.9% NaCl solution after surgery), empty adenovirus group, recombinant Galectin-9 adenovirus group, cardiac graft survival At the time of transplantation, the spleen was removed at 14 days after cardiac arrest or transplantation, and mononuclear cells were isolated. The proportion of CD4 + CD25 + Treg cells was detected by flow cytometry. The levels of IL-10 and TGF-β1 in peripheral blood were detected by ELISA. Results Compared with control group and empty adenovirus group, the survival time of cardiac grafts in recombinant Galectin-9 adenovirus group was significantly increased (P <0.01), and the proportion of CD4 + CD25 + Treg cells in recombinant Galectin-9 adenovirus group was significantly increased (P <0.01). The levels of IL-10 and TGF-β1 in recombinant Galectin-9 adenovirus group were significantly higher than those in control group and empty adenovirus group (P <0.01). Conclusion Recombinant Galectin-9 adenovirus can promote the differentiation and proliferation of CD4 + CD25 + Treg cells in vivo and significantly improve the survival time of cardiac allografts in mice.
