丹那唑(danazol)为合成的乙炔睾丸酮衍生物,临床上用来治疗子宫内膜异位、乳腺发育不良和性早熟,也有用作节育药。但其作用机制尚不完全明了。作者等以前的研究表明丹那唑通过对下丘脑雄激素受体的作用抑制促性腺激素。为确定丹那唑对性腺的直接作用,作者等又进行了下列研究: 1.对月经周期黄体期的影响:10只性成熟的有规则月经的雌性恒河猴,分别单独置于笼内,根据血浆雌激素水平测定和腹腔检查确定其排卵日期。实验包括3个连续的月经周期,第一个周期为对照周期,第二个为用药周期,第三个为停药周期,于第二周期从排卵后第一天起至月经开始每天管饲丹那唑或乳糖400mg(各5只动物)。每周期从第八天起至月经开始每天上午肌注氯胺酮使动物镇静后取股静脉血2~3ml,用放射免疫法测定血中FSH、
Danazol is a synthetic acetylene testosterone derivative that is clinically used to treat endometriosis, dysplasia and precocious puberty and is also used as a birth control pill. But its mechanism of action is not completely understood. Previous studies by the authors showed that danazol suppresses gonadotropins by acting on hypothalamic androgen receptors. To determine the direct effects of Danazol on the gonads, the authors conducted the following additional studies: 1. Effects on the Luteal Phase of the Menstrual Cycle: Ten sexually mature female rhesus macaques with regular menstruation were placed individually in cages, Plasma estrogen levels and abdominal examination to determine their ovulation date. The experiment consisted of three consecutive menstrual cycles. The first cycle was the control cycle, the second cycle was medication, and the third cycle was withdrawal.
