目的:选择一种比色法作为白花蛇舌草总黄酮的最优测定方法。方法:以芦丁为标准品,通过直接测定法、硝酸铝法、氯化铝法3种检测方法的比较,确定一种检测方法作为白花蛇舌草总黄酮的测定最优方法。结果:经过比较,选择氯化铝法作为测定白花蛇舌草总黄酮含量更好,在410nm波长下测定,样品在0.05mg~0.3mg范围内,其浓度与吸光度呈良好的线性关系,相关系数R=0.9997,平均加样回收率为99.84%,RSD=1.31%(n=5),稳定性、精密度和重现性均理想。结论:氯化铝显色法的大部分指标都好于其他两种比色法,且分析时间较短,准确度高,因此三氯化铝比色法更适合白花蛇舌草总黄酮含量的测定。
Objective: To choose a colorimetric method as the optimal method for determining the total flavonoids of Hedyotis diffusa. Methods: Rutin was used as a standard sample to determine the best method for the determination of total flavonoids in Hedyotis diffusa Willd. By direct comparison, comparison between the three detection methods of aluminum nitrate and aluminum chloride. Results: After comparison, the method of aluminum chloride was selected to determine the content of total flavonoids in Hedyotis diffusa Willd. Better at 410nm wavelength. The range of 0.05mg ~ 0.3mg was linear with the absorbance. The correlation coefficient R = 0.9997, the average recovery was 99.84%, RSD = 1.31% (n = 5), the stability, precision and reproducibility were ideal. Conclusion: Most of the indicators of aluminum chloride colorimetric method are better than the other two colorimetric method, and the analysis time is short, the accuracy is high, so aluminum trichloride colorimetric method is more suitable for the total flavonoid content of Hedyotis diffusa Willd Determination.
