以松茸子实体为DNA参照样品,对疑似松茸菌株Z-s、R-b、H-c进行亲菌鉴定。运用RAPD-PCR技术,采用筛选的5个随机引物对1个供试松茸子实体及3个疑似松茸菌株Z-s、R-b、H-c进行RAPD(Random Amplified Polymorphic) -PCR鉴定,获得DNA指纹图讲;分析菌株Z-s、R-b、H-c间及其与松茸子实体间DNA同源性及遗传相似度,以鉴定疑似松茸菌株Z-s、R-b、H-c是否是目的松茸菌株。实验结果其相似系效分别为:Ssz 35.7%;Sbz 74.1%;Scz 76.9%,可以看出疑似菌株Z-s与松茸子实体DNA指纹图谱差异比较明显,相似系数只有Ssz 35.7%,而疑似菌株R-b和H-c则和松茸子实体的DNA指纹图谱比较相近,相似系数分别为Sbz 74.1%,Scz 76.9%。在分离菌株的形态及生物学特征进行表型鉴别的基础上,初步判定疑似菌株R-b和H-c-很可能是目的松茸菌株。
Tricholoma matsutake strains were used as DNA reference samples for the identification of the suspected Matsutake strains Z-s, R-b and H-c. RAPD-PCR was used to screen DNA fingerprinting with RAPD (Random Amplified Polymorphic) -PCR for one Tricholoma matsutake seed body and three suspected Matsutake strains Zs, Rb and Hc using five random primers. Strains Zs, Rb, Hc and their matsutake between the entities and DNA similarity and genetic similarity in order to identify the suspected matsutake strains Zs, Rb, Hc is the purpose of matsutake strains. The similarity of the experimental results were: Ssz 35.7%; Sbz 74.1%; Scz 76.9%. It can be seen that there is a significant difference in the DNA fingerprinting between the suspect strain Zs and Tricholoma matsutake subsp. The similarity coefficient is only 35.7% of Ssz, while the suspected strains Rb and Hc and Matsuzaka fruiting bodies DNA fingerprinting similar, similar coefficients were 74.1% Sbz, Scz 76.9%. Based on the phenotypic identification of the morphological and biological characteristics of isolates, it was initially determined that the suspected strains R-b and H-c- are likely to be Matsutake mushrooms.
