目的:探讨消瘀泄浊饮对UUO模型大鼠的保护作用及作用机制。方法:实验采用UUO大鼠模型,将32只SD大鼠随机分为假手术组(S组)、模型组(M组)、洛汀新治疗组(B组)、消瘀泄浊饮组(X组)。分组用药并监测体重,给药14天后处死大鼠。测定大鼠左肾指数,血清肌酐、尿素氮、白蛋白水平;肾组织HE染色观察病理改变;用免疫组织化学方法检测肾组织α-SMA的表达。结果:消瘀泄浊饮能够明显降低实验大鼠血清肌酐、尿素氮水平,升高白蛋白水平,能降低肾组织中α-SMA蛋白表达,与模型组比较有显著性差异,形态学观察也显示其肾脏损害轻于模型组。结论:消瘀泄浊饮能有效地延缓肾间质纤维化的进程,可通过抑制α-SMA的表达,减少肌成纤维细胞的活化和形成,从而起到抗肾纤维化的作用。
Objective: To investigate the protective effect of Xiaoyu Xiezhuo Yin on UUO model rats and its mechanism. Methods: Thirty-two Sprague-Dawley rats were randomly divided into sham operation group (model group), model group (M group), Lotensin treatment group (group B), Xiaoyuxiaozhuo Decoction group X group). The animals were divided into groups and monitored for body weight. Rats were sacrificed 14 days after administration. The left renal index, serum creatinine, urea nitrogen and albumin were determined. The pathological changes of renal tissue were observed by HE staining. The expression of α-SMA in renal tissue was detected by immunohistochemical method. Results: Xiaoyu Xiezhuo Yin can significantly reduce serum creatinine and urea nitrogen levels, increase albumin level in renal tissue and decrease the expression of α-SMA protein in renal tissue, which is significantly different from the model group. Morphological observation It showed that kidney damage was lighter than model group. Conclusion: Xiaoyu Xiezhuo Decoction can effectively delay the process of renal interstitial fibrosis, which can prevent renal fibrosis by inhibiting the expression of α-SMA and reducing the activation and formation of myofibroblasts.
