目的:探讨黄精多糖对去卵巢大鼠骨微结构、Ca、P、OPG和RANKL蛋白表达的影响。方法:5月龄雌性大鼠30只,随机分5组:假手术(Sham)组,去势(OVX)组,黄精多糖高、中、低(H-,M-,L-PSP)组。Sham组、OVX组灌胃生理盐水,PSP组灌胃黄精多糖。8周后,观察大鼠胫骨显微结构,Ca、P、OPG和RANKL蛋白表达。结果:OVX组体重、Ca、P、RANKL、骨小梁Tb.Sp较Sham组升高,骨小梁BV/TV、Tb.Th、Tb.N和OPG降低;H-,M-PSP组体重、PSP组Ca和H-PSP组P、H-PSP组RANKL、PSP组Tb.Sp较OVX组降低,H-,M-PSP组OPG和PSP组BV/TV、Tb.Th、Tb.N升高;差异具有统计学意义(P<0.05)。结论:黄精多糖对骨质疏松防治主要通过改善骨微结构、降低骨转换、提高OPG蛋白和降低RANKL蛋白表达。
Objective: To investigate the effects of Polygonatum Polysaccharides (PNS) on the microarchitecture, Ca, P, OPG and RANKL protein expression in ovariectomized rats. Methods: Thirty five-month-old female rats were randomly divided into five groups: Sham group, OVX group, H-, M- and L-PSP groups. Sham group, OVX group were given normal saline, PSP group was given Polygonatum polysaccharide. After 8 weeks, the tibia microstructure, Ca, P, OPG and RANKL protein expression in rats were observed. Results: The body weight, Ca, P, RANKL, TB.Sp of trabecular in OVX group were higher than those in Sham group, BV / TV, Tb.Th, Tb.N and OPG of trabecular were decreased; , Tb.Sp in PSP and H-PSP groups were lower than those in OVX group, but BV / TV, Tb.Th and Tb.N in OPG and PSP groups in H- and M-PSP group were significantly increased High; the difference was statistically significant (P <0.05). Conclusion: Polygonatum polysaccharide can prevent and cure osteoporosis mainly by improving the microstructure of bone, reducing bone turnover, increasing OPG protein and decreasing RANKL protein expression.
