目的:研究肺癌组织中上皮钙粘蛋白(E-CD)基因启动子区5′CpG岛甲基化状况,揭示肺癌发生、发展过程中的分子水平变化和可能作用机制。方法:应用甲基化特异性PCR(methylation specific PCR)检测41例肺癌手术标本及同一病例正常肺组织中E-CD基因启动子区5′CpG岛甲基化水平。结果:E-CD基因启动子区甲基化阳性率在肺癌中显著高于正常肺组织,与肺癌的分型、细胞分化程度、淋巴结转移、TNM分期明显相关。结论:E-CD基因启动子区甲基化水平可作为较早期预测肺癌浸润转移程度,评价预后的有效指标。
Objective: To investigate the 5’CpG island methylation status of E-cadherin gene promoter region in lung cancer and to reveal the molecular changes and possible mechanism of lung cancer carcinogenesis and development. Methods: methylation-specific PCR (methylation specific PCR) was used to detect methylation status of 5’CpG island in promoter region of E-CD gene in 41 lung cancer specimens and normal lung tissues from the same patient. Results: The positive rate of E-CD gene promoter methylation in lung cancer was significantly higher than that in normal lung tissue, which was significantly correlated with the type of lung cancer, cell differentiation, lymph node metastasis and TNM stage. CONCLUSION: The methylation level of E-CD gene promoter region can be used as an effective indicator to predict the degree of invasion and metastasis of lung cancer in the early stage.
