目的探讨4-HPR联合顺铂对人卵巢癌细胞株SKOV3增殖和Akt/Bax表达的影响。方法 MTT法检测单独使用4-HPR、顺铂及联合使用对SKOV3细胞增殖的影响;用流式细胞仪检测4-HPR、顺铂对SKOV3细胞周期和Akt/Bax表达的影响。结果顺铂联合不同浓度的4-HPR可以显著抑制SKOV3细胞的增殖,同单独使用顺铂相比抑制作用较强,差异显著(P<0.05)。顺铂联合4-HPR同使用顺铂相比可以使G0~G1期细胞比例进一步下降,S期和G2~M期细胞比例进一步上升,差异显著(P<0.01)。顺铂联合4-HPR的凋亡率同单独用顺铂组相比差异有统计学意义(P<0.01)。顺铂联合不同浓度的4-HPR时可以使SKOV3细胞Akt的表达下调和促进Bax的表达的上调,同单独使用顺铂相比差异有统计学意义(P<0.01)。结论顺铂联合4-HPR可以更好地抑制细胞株SKOV3增殖,使得细胞停滞在S期和G2~M期,这可能与可以促进Akt的表达下调和Bax的表达上调有关。
Objective To investigate the effects of 4-HPR combined with cisplatin on the proliferation and Akt / Bax expression of human ovarian cancer cell line SKOV3. Methods MTT assay was used to detect the effect of 4-HPR and cisplatin on the proliferation of SKOV3 cells. The effects of 4-HPR and cisplatin on the cell cycle and Akt / Bax expression were examined by flow cytometry. Results cisplatin combined with different concentrations of 4-HPR can significantly inhibit the proliferation of SKOV3 cells, compared with cisplatin alone, the inhibition is stronger, the difference was significant (P <0.05). Compared with cisplatin, cisplatin combined with 4-HPR could further decrease the proportion of cells in G0-G1 phase and increase the proportion of cells in S phase and G2-M phase (P <0.01). The apoptosis rate of cisplatin combined with 4-HPR was significantly lower than that of cisplatin alone (P <0.01). Cisplatin combined with different concentrations of 4-HPR can down-regulate the expression of Akt and up-regulate the expression of Bax in SKOV3 cells, which is significantly different from cisplatin alone (P <0.01). Conclusion Cisplatin combined with 4-HPR can inhibit the proliferation of cell line SKOV3 and arrest cells in S phase and G2 ~ M phase, which may be related to down-regulation of Akt and up-regulation of Bax.
