目的探讨大黄素对兔主动脉平滑肌细胞(CCC-SMC-1)凋亡基因Fas mRNA表达的影响。方法 MTT法观察大黄素对兔主动脉平滑肌细胞增殖的抑制作用,RT-PCR法检测大黄素对兔主动脉平滑肌细胞中Fas mRNA的表达。结果MTT法显示大黄素在0.05~0.25 mmol·L~(-1)浓度范围内对CCC-SMC-1细胞增殖有抑制作用,作用24 h后的IC_(50)为0.10 mmol·L~(-1),且IC_(50)随作用时间的延长显著前移。RT-PCR法显示,不同剂量的大黄素作用于兔主动脉平滑肌细胞24 h后,与正常组细胞相比,Fas mRNA的表达水平均升高(P<0.05)。结论大黄素能抑制CCC-SMC-1细胞的增殖,其机制可能与上调的Fas mRNA表达有关。
Objective To investigate the effect of emodin on the expression of Fas mRNA in aortic smooth muscle cells (CCC-SMC-1). Methods MTT assay was used to observe the inhibitory effect of emodin on the proliferation of rabbit aortic smooth muscle cells. The expression of Fas mRNA was detected by RT-PCR in rabbit aortic smooth muscle cells. Results MTT assay showed that emodin could inhibit the proliferation of CCC-SMC-1 cells in the concentration range of 0.05-0.25 mmol·L -1, and the IC 50 after 24 h treatment was 0.10 mmol·L -1 1), and IC_ (50) with the extension of the role of time significantly advanced. RT-PCR showed that the expression of Fas mRNA was significantly increased in emodin-treated rabbit aorta smooth muscle cells for 24 h (P <0.05) compared with normal cells. Conclusion Emodin can inhibit the proliferation of CCC-SMC-1 cells, which may be related to the up-regulated expression of Fas mRNA.
