用20只SD大鼠的坐骨神经片段培养在涂有层粘蛋白的聚酯纤维上,对照组培养在无层粘蛋白的聚酯纤维上.培液由20%小牛血清和80%DMEM组成,37℃,5%二氧化碳培养箱培养2周.在培养24,48,72小时,观察雪旺细胞迁移情况,测量迁移最远的雪旺细胞离组织块边缘的距离.结果显示:实验组和对照组相比较,雪旺细胞从组织中迁出的时间早.速度快,数量多.在体外培养条件下.显示了层粘蛋白对雪旺细胞在三维网架上迁移的促进作用.对层粘蛋白促进雪旺细胞迁移的影响和作用机制作了讨论,并探讨其在神经移植中的应用前景.
Sciatic nerve segments of 20 SD rats were cultured on laminin-coated polyester fibers and control animals on laminin-free polyester fibers.The broth consisted of 20% calf serum and 80% DMEM, 37 ℃, 5% carbon dioxide incubator for 2 weeks.After culturing for 24, 48 and 72 hours, the migration of Schwann cells was observed and the distance between the most distant Schwann cells migrated from the edge of the tissue was measured.The results showed that the experimental group and the control In contrast, Schwann cells migrated out of tissues earlier, faster, and more in quantity, demonstrating the promoting effect of laminin on the migration of Schwann cells on the three-dimensional network in vitro.
