目的研究自然早产胎盘中细胞凋亡抑制因子survivin表达水平。方法收集我院2014年8月~2015年8月收治的60例胎盘,按随机表法将其平均分为两组,病例数均为30例,其中足月产分娩的胎盘为对照组,早产分娩的胎盘为观察组,对两组分别实施RT-PCR法(survivin m RNA定量测定)和细胞凋亡抑制因子survivin免疫检测。利用免疫组化检测的方法对两组胎盘组织间survivin因子的差别进行比较,并进一步研究其与survivin m RNA之间的定量关系,阐明Survivin基因表达在自然早产发生中的意义。结果经统计学分析,结果如下:两组患者的基本资料(主要包括产妇年龄、孕周和胎儿出生时的体重等)之间的比较差异均不具有显著性(P>0.05);观察组的Survivin蛋白表达比例低于对照组,两者之间的差异具有显著性(χ2=12.123,P<0.05);观察组的Survivin蛋白表达量低于对照组,两者之间的差异具有显著性(t=2.134,P<0.05)。观察组的Survivin m RNA的表达比例低于对照组,两者之间的差异具有显著性(χ2==11.321,P<0.05)。观察组Survivin蛋白(△CT值)低于对照组,两者之间的差异具有显著性(t=2.235,P<0.05)。结论在胎盘滋养细胞中,凋亡抑制因子survivin的表达水平下降会促使细胞过度凋亡现象的发生,这个机制又会致使胎盘功能下降,进而发生早产。
Objective To study the expression of survivin in the placenta of naturally premature placenta. Methods Sixty cases of placenta collected from August 2014 to August 2015 in our hospital were divided into two groups according to the random table method. The number of cases were all 30 cases. The placenta of full-term delivery was the control group, The placenta of childbirth was the observation group, and the two groups were detected by RT-PCR (quantitative determination of survivin m RNA) and survivin immuno-detection of apoptosis inhibitor. The difference of survivin factor between the two groups of placenta was compared by immunohistochemistry and the relationship between survivin mRNA and survivin m RNA was further studied to clarify the significance of Survivin gene expression in natural preterm birth. The results of statistical analysis, the results are as follows: The two groups of patients with basic information (including maternal age, gestational age and birth weight, etc.) were not significantly different between the differences (P> 0.05) Survivin protein expression was lower than the control group, the difference was significant (χ2 = 12.123, P <0.05); Survivin protein expression in the observation group was lower than the control group, the difference between the two groups was significant t = 2.134, P <0.05). The expression of Survivin m RNA in the observation group was lower than that in the control group, and the difference was significant (χ2 == 11.321, P <0.05). Survivin protein (△ CT value) in the observation group was lower than that in the control group, the difference was significant (t = 2.235, P <0.05). Conclusions The decrease of the expression of survivin in placental trophoblast cells will lead to the phenomenon of excessive apoptosis of cells. This mechanism will lead to the decrease of placental function and premature delivery.
