目的:用表面增强激光解吸离子化飞行时间质谱(SELDI-TOF-MS)技术建立喉癌患者血清蛋白质组构型,并筛选出相对特异标记物。方法:收集32例喉癌患者术前、15例喉癌患者术后10 d及40例正常人的血清,用固定金属亲和表面蛋白质芯片(IMAC-Cu)和SELDI-TOF-MS技术检测蛋白质谱的表达。获得的蛋白质谱采用Ciphergen公司的Biomarker Wizard和Biomarker Pattern软件分析。结果:通过对喉癌患者术前、术后10 d与正常人血清蛋白质谱分析发现,共有26个蛋白质的表达量有明显差异。并获得平均相对分子质量分别为4 048.09、5 073.79、5 339.29、3 795.38、9 187.24、15 323.8、6 111.92、6 943.87这8个蛋白质组成的模板,可将喉癌术前、术后及对照组正确分组。其正确分组率分别为71.88%(23/32)、66.67%(10/15)、75%(30/40)。结论:SELDI-TOF-MS技术是一种快速、简便易行且高通量的分析方法,能直接筛选出喉癌患者血清中相对特异的潜在标记物,且可能具有较好的临床应用前景。
OBJECTIVE: To establish the serum proteome configuration of laryngeal carcinoma patients by surface enhanced laser desorption / ionization time of flight mass spectrometry (SELDI-TOF-MS) and to screen out specific markers. Methods: Serum samples from 32 patients with laryngeal cancer before operation and 15 patients with laryngeal cancer were collected at 10 days and 40 normal controls. IMAC-Cu and SELDI-TOF-MS were used to detect the protein Expression of spectrum. The obtained protein profiles were analyzed using Biomarker Wizard and Biomarker Pattern software from Ciphergen. Results: According to the serum protein profiles of preoperative, postoperative 10 and postoperative laryngeal cancer patients, the expression of 26 proteins was significantly different. And obtained the average relative molecular mass of 4 048.09,5 073.79,5 339.29,3 795.38,9 187.24,15332.8,6 111.92,6 943.87 the template of the eight proteins can laryngeal cancer preoperative and postoperative and control Group correctly grouped. The correct grouping rates were 71.88% (23/32), 66.67% (10/15) and 75% (30/40), respectively. Conclusion: SELDI-TOF-MS is a rapid, simple and easy-to-use high-throughput analytical method that can directly screen the relative markers of serum specific in laryngeal cancer patients and may have a good clinical application prospect.
