目的:研究小鼠胸膜间皮细胞(pleural mesothelial cell,PMC)免疫相关基因表达谱的状况。从而探索用现代免疫及基因芯片技术评价解释中医藏象理论的新方法。方法:取8只BALB/c小鼠胸膜组织块剪切成碎块,提取mRNA,经标记、杂交、清洗、芯片扫描、图象采集与数据分析等过程获得基因表达谱。结果:共筛选出小鼠胸膜间皮细胞免疫相关表达基因204条。其中主要有免疫应答及调节基因56条;炎性反应基因32条;非特异性免疫基因14条;T细胞增殖、活化、调节等相关基因23条,B细胞活化、分化、调节等相关基因9条;急性反应期基因5条;抗原加工与提呈基因4条;Ⅱ型超敏反应正向调节基因5条。结论:胸膜间皮细胞既表达免疫相关的正向调节基因亦表达负向免疫相关的调节基因。提示胸膜可能为一免疫协调器官,这对丰富中医藏象理论与肺气的实质将提供分子生物学的基础研究依据与新的思路。
Objective: To investigate the expression of immune related gene in mouse pleural mesothelial cell (PMC). In order to explore the use of modern immunization and gene chip technology to evaluate the interpretation of the theory of Tibetan medicine, a new method. Methods: Eight BALB / c mice were cut into pieces and cut into pieces. The mRNA expression was obtained by labeling, hybridization, washing, chip scanning, image acquisition and data analysis. Results: A total of 204 mouse pleuropneumoniae immune-related genes were screened out. Among them, there are 56 immune response and regulation genes, 32 inflammatory response genes, 14 non-specific immune genes, 23 T cell proliferation, activation and regulation related genes, 9 B cell activation, differentiation and regulation related genes ; 5 acute response genes; 4 antigen processing and presenting genes; and 2 positive response genes of type 2 hypersensitivity. CONCLUSION: Pleural mesothelial cells express both immune-related up-regulated genes and down-regulated immune-related regulatory genes. Suggesting that the pleura may be an immune coordination organ, which enrich the theory of TCM and lung essence will provide the basis of molecular biology and new ideas.
