目的:检测并鉴定感染组织中的克里米亚-刚果出血热病毒。方法:分别经皮下与腹腔两种途径感染乳小鼠;应用RT-PCR技术对病毒核酸进行检测;利用透射电镜超薄切片技术对病毒进行形态学鉴定。结果:RT-PCR可在心脏、肝脏、脾脏、肾脏以及脑组织内检测到病毒核酸,透射电镜下可在肝细胞胞浆内观察到形态典型的病毒粒子。结论:应用RT-PCR技术可以对感染组织中的克里米亚-刚果出血热病毒进行检测,利用透射电镜可以对病毒的形态进行观察与鉴定。
Purpose: To detect and identify the Crimean-Congo hemorrhagic fever virus in infected tissues. Methods: The mice were infected by both subcutaneous and intraperitoneal routes respectively. The viral nucleic acid was detected by RT-PCR. The morphology of the virus was identified by transmission electron microscopy. Results: RT-PCR could detect the virus nucleic acid in the heart, liver, spleen, kidney and brain tissue. The typical virus particles were observed in the cytoplasm of hepatocytes under transmission electron microscope. CONCLUSION: The Crimean-Congo hemorrhagic fever virus in infected tissues can be detected by RT-PCR. The morphology of the virus can be observed and identified by transmission electron microscopy.
