目的:探究管萼山豆根中生物碱的薄层色谱(TLC)分析方法,并以此法制备其中的两种生物碱。方法:对管萼山豆根氯仿层总生物碱的TLC展开剂及显色方法等进行研究,采用制备薄层色谱(PTLC)法制备苦参碱和氧化苦参碱,并用HPLC检测。结果:TLC展开剂为:V(石油醚)∶V(氯仿)∶V(甲醇)=5∶3∶2(氨水饱和0.5h),碘蒸气显色,斑点清晰,不拖尾。用PTLC制备得到苦参碱和氧化苦参碱。结论:建立了管萼山豆根中生物碱的TLC快速分析分离方法,不仅可作为检测、分离及药材鉴定的依据,而且可为柱层析的洗脱条件提供参考。
Objective: To investigate the TLC method of alkaloids in the root of Radix et Rhizoma. And to prepare two kinds of alkaloids by this method. Methods: TLC development and colorimetric methods of the total alkaloids of C. epidermis and R. solanum were studied. Matrine and oxymatrine were prepared by preparative thin layer chromatography (PTLC) and detected by HPLC. Results: The developing solvent of TLC was V (petroleum ether): V (chloroform): V (methanol) = 5: 3: 2 (saturated ammonia water for 0.5h). Matrine and oxymatrine were prepared using PTLC. Conclusion: The rapid TLC method for the determination of alkaloids in the roots of C. albiflorus was established. It can not only be used as the basis for the detection, separation and identification of medicinal materials, but also provide a reference for the elution conditions of column chromatography.
