目的探索肿瘤抑制基因p53、p16变异在脑肿瘤发病中的意义。方法对23例脑肿瘤人病原发肿瘤标本用免疫组化LSAB法行肿瘤抑制基因p53、p16蛋白检测。结果星形细胞瘤Ⅰ级9例,p16蛋白4例表达30%以下,5例不表达,p53蛋白3例表达30%以下,6例不表达;Ⅱ~Ⅲ级7例;p16蛋白1例表达30%以下,6例不表达,p53蛋白4例表达30%~60%,3例不表达;脑膜瘤、垂体瘤7例,p16蛋白3例表达30%以下,4例不表达,p53蛋白2例表达30%以下,5例不表达。结论p53、p16蛋白缺失、变异是肿瘤发生的重要的原因之一,p53变异主要见于恶性胶质细胞瘤,偏良性的胶质细胞瘤和脑膜瘤、垂体瘤,p53蛋白轻度阳性表达,提示肿瘤可能处于早期恶性变过程中。p16蛋白缺失多见于恶性胶质瘤,也见于部分良性肿瘤。
Objective To explore the significance of the mutation of tumor suppressor gene p53 and p16 in the pathogenesis of brain tumor. Methods Twenty - three cases of human brain tumor with primary tumor were examined by immunohistochemical method of LSAB with tumor suppressor genes p53 and p16. Results There were 9 cases with grade Ⅰ in astrocytoma, 30 cases with p16 protein in 4 cases, 5 cases without expression, 3 cases with p53 protein less than 30%, 6 cases without expression, 7 cases with Ⅱ ~ Ⅲ grade and 1 case with p16 protein Up to 30%, 6 cases did not express, 4 cases expressed 30% -60% p53 protein, 3 cases did not express; 7 cases of meningioma and pituitary tumor, 3 cases of p16 protein expressed less than 30%, 4 cases did not express p53 protein 2 Cases of expression below 30%, 5 cases do not express. Conclusions The deletion and mutation of p53 and p16 proteins are one of the important causes of tumorigenesis. The variation of p53 is mainly found in malignant glioma, benign glioma and meningioma, pituitary tumor and mild positive expression of p53 protein Tumor may be in the process of early malignant transformation. Deletion of p16 protein is more common in malignant gliomas, also found in some benign tumors.
